茵陈对肝内胆汁淤积湿热证大鼠利胆退黄作用机制的研究

目的通过研究茵陈对肝内胆汁淤积湿热证大鼠肝组织中胆盐输出泵(BSEP)基因表达的影响,从分子水平探讨茵陈利胆退黄的作用机制。方法 45只雄性SD大鼠随机分为3组:正常组、模型对照组和茵陈组。检测治疗后大鼠血清生化指标、肝脏组织病理的变化和肝脏BSEP的表达。结果治疗后茵陈组大鼠体质量增加明显高于模型对照组(P<0.05)。茵陈组大鼠血清TB、DB、ALT、AST、ALP、TBA浓度低于模型对照组(P<0.05);HE染色显示茵陈组大鼠肝脏的病理损害轻于模型对照组。免疫组织化学染色显示茵陈组大鼠比模型对照组有更多的BSEP阳性细胞。荧光定量RT-PCR法显示模型对照组大鼠肝组织中BSEP mRNA表达明显低于茵陈组(P<0.05)。结论茵陈能有效改善肝内胆汁淤积湿热证大鼠的肝功能、减轻病理损害,并能上调肝脏BSEP的表达。

The Choleretic and Jaundice-relieving Mechanism of Yinchen on Rat with Intrahepatic Cholestasis and Damp-heat

Objective To investigate the choleretic and jaundice-relieving mechanism of Yinchen,the BSEP expression levels in the livers of rats with intrahepatic cholestasis and damp-heat were detected after treatment with Yinchen. Methods Forty-five male SD rats were randomly assigned into 3 groups: normal,model control and Yinchen groups.The body weights,liver functions,liver pathologies and BSEP expressions in rat livers were detected. Results The rat weight gain values in Yinchen group were greater than that of model control group after treatment(P<0.05).The blood serum levels of TB,DB,ALT,AST,ALP and TBA in Yinchen group were lower than those in model control group(P<0.05).The degrees of pathological damage of rat hepatocytes in Yinchen group were weaker than those in model control group as shown in the hematoxylin-eosin staining.More BSEP postive expression hepatocytes were found in rats of Yinchen group than those of model control group as shown in the immunohistochemical staining.In the analysis of fluorescent quantitative RT-PCR,the BSEP mRNA expression levels of rat hepatocytes in Yinchen group were higher than those of model control group(P<0.05). Conclusion Yinchen can improve the liver function,mitigate the pathological damage and increase the BSEP expression level in the liver of rat with intrahepatic cholestasis and damp-heat.
