燕窝中唾液酸含量测定方法的研究

摘    要:
目的研究燕窝中唾液酸含量的测定方法,制定燕窝的质量标准。方法燕窝经过磷酸水解,其中的唾液酸(即N-乙酰神经氨酸)从唾液酸糖蛋白的结合状态游离出来,用高效液相色谱法进行检测,以唾液酸含量作为评价指标,通过三因素三水平正交试验,优选出最佳的水解条件。结果唾液酸浓度在20~330μg/mL范围内线性良好,平均回收率为98.4%,RSD为1.02%(n=6)。结论此法具有稳定性及重复性好、灵敏度高、操作简单等特点,能够准确地检测出燕窝中的唾液酸含量。

Abstract:
Purpose To study on the determination method of the content of sialic acid in edible bird′s nest(EBN),so as to establish EBN quality standard.Methods After acid hydrolysis,the sialic acid in the bird′s nest(that is N-Acetylneuraminic acid)is free from the combination of the state of glycoprotein sialic acid.High performance liquid chromatography.Is used to detect the sialic acid as evaluation index.The optimized conditions are found on the basis of orthogonal experiments of three factors and three levels.Results The linear range of sialic acid was 20-330 μg/mL.The average recovery was 98.4% with RSD of 1.02%(n=6).Conclusion This method has the advantages of good stability,high sensitivity,simple operation,etc.It can accurately detect the content of the sialic acid in bird′s nest.
