摘 要: | 目的探讨防御素(HNP1-3)和白细胞介素-8(IL-8)在大肠湿热型溃疡性结肠炎(UC)结肠组织中的表达分布及两者之间的相关性,观察中药溃结灵煎剂对HNP1-3和IL-8的影响,探讨其作用机制。方法大肠湿热型活动期UC患者随机分为2组,治疗组予溃结灵+柳氮磺胺吡啶治疗,对照组予柳氮磺胺吡啶,疗程为12个月。采用常规免疫组化S-P法检测治疗前后肠黏膜组织内HNP1-3及IL-8的蛋白表达情况。结果随着病变组分级程度的增高,HNP1-3、IL-8细胞因子的表达逐渐增强,两者呈明显的正相关。治疗组和对照组治疗后HNP1-3、IL-8表达较治疗前明显减少(P0.01,P0.05)。2组HNP1-3、IL-8的蛋白表达治疗后差值比较,差异有统计学意义(P0.05)。结论溃结灵配合柳氮磺胺吡啶治疗很可能是通过抑制IL-8等细胞因子对中性粒细胞的激活,不能或减少释放细胞毒性防御素,使损伤的内皮细胞得以修复,继而达到治疗UC的作用。

Abstract: | Objective To probe into the expression of HNP1-3 and IL-8 in colon tissue of ulcerative colitis (UC) of damp-heat in large intestine stagnation type and their relationship, to observe the effect of Kuijieling on HNP1-3 and IL-8 and study its intervention effect in treating UC. Methods The patients of active UC (damp-heat syndrome in large intestine stagnation type) were randomly divided into treatment group and control group, and treated with Kuijieling + SASP and SASP respectively. The protein expressions of HNP1-3 and IL-8 in all cases were detected by S-P immunohistochemical method. Results The level of HNP1-3 and IL-8 in colon mueosa of UC patients were much higher than that of normal controls. The expression of HNP1-3 were significantly correlated with that of IL-8. In the treatment group, the expression of HNP1-3 and IL-8 were decreased by treatment (P〈0.05). The differences in HNP1-3 and IL-8 between the two groups were significant after treatment (P〈0.05). Conclusion Kuijieling and SASP has the function of restoring injured endothelial cell by restraining IL-8 arousing neutrocyte and reducing HNP1-3, so as to treating UC. |
