摘 要: | 目的研究芹菜素对小鼠睾丸生殖细胞周期的影响。方法将50只SPF级健康成年雄性昆明小鼠随机分为阴性对照组(生理盐水)、溶剂对照组(0.5%DMSO)和低(84mg/kg)、中(168mg/kg)、高(252mg/kg)剂量芹菜素染毒组,每组10只。采用灌胃方式进行染毒,染毒剂量为10ml/kg,每天1次,连续灌胃14天,于末次灌胃后第21天处死小鼠,分别称重附睾、睾丸,并计算脏器系数。采用流式细胞仪检测小鼠生精细胞的周期。结果染毒前后各组小鼠体重间比较,差异均无统计学意义(P0.05)。除高剂量芹菜素染毒组小鼠附睾湿重高于中剂量芹菜素染毒组(P0.05)外,各组小鼠睾丸、附睾湿重及其脏器系数间比较,差异均无统计学意义(P0.05)。细胞周期分析结果显示,与阴性对照组比较,低、高剂量芹菜素染毒组小鼠睾丸组织中的二倍体细胞所占百分比下降,低剂量芹菜素灌胃可引起G0/G1期细胞明显降低,低、高剂量芹菜素染毒组小鼠睾丸组织1C∶2C值明显增加,差异均有统计学意义(P0.05)。结论芹菜素对小鼠生精细胞有一定的抑制作用。

Effect of Apigenin on Cell Cycle of Spermatogenic Cells of Male Mice |

Abstract: | Objective To explore the effect of apigenin on cell cycle of spermatogenic cells of male mice. Methods Fifty male Kunming mice of SPF were randomly divided into 5 groups according to body weight (negative control group, solvent control group,low-dose group of apigenin, moderate-dose group of apigenin, high-dose group of apigenin), 10 in each. Apigenin was given through gavage, once a day, for 14 consecutive days. After 21 days of last gavage,all mice were sacrificed. The weight of testis and epididymis was measured and cell cycle of spermatogenic cells were detected by flow cytometry. Results There was no significant difference in weight after and before the treatment (P>0.05). Except that the wet weight of the epididymis in high-dose group of apigenin was significantly different compared with middle-dose group of apigenin (P<0.O5),there was no statistical difference among the others groups between wet weight and organ coefficient of testis and epididymis (P>0.05). Cell cycle analysis showed that compared with the negative control group, the percent of diploid cells in low and high-dose group was decreased, the proportion of G_0/G_1 phase cells in low-dose group of apigenin was reduced(P<0.05); IC:2C was increased (P<0.05). Conclusion Apigenin has a some adverse effects on spermatogenic cells in male mice. |
