哮喘小鼠肺组织中水通道蛋白5和黏蛋白5AC的表达变化及其相关性研究

摘    要:目的探讨支气管哮喘(以下简称哮喘)小鼠肺组织水通道蛋白5(AQP5)和黏蛋白MUC5AC基因和蛋白的表达变化及其两者的相关性。方法制作小鼠哮喘模型,测定支气管肺泡灌洗液细胞分类及肺组织湿干重比值,应用Real-time PCR法测定哮喘组(OVA组)和对照组肺组织中AQP5和MUC5AC mRNA的表达,应用免疫组化法测定AQP5在肺组织中的分布,免疫印记法、ELISA法测定AQP5和MUC5AC蛋白在肺组织中的表达。结果小鼠哮喘组肺组织中AQP5表达较对照组明显减低(P<0.01),而MUC5AC表达显著升高(P<0.01),两者呈负相关(r=0.901,P<0.01)。结论哮喘时肺组织中AQP5表达的减低可能与气道MUC5AC表达的升高有关,从而促进了气道黏液过度分泌。

Abstract:[Objective] To investigate the expression of aquaporin5 and MUC5AC in lung in rat asthma model and analyse the relationship between them. [Methods] After making asthmatic murine model, expression and their distribution of AQP5 and MUC5AC in lung were detected by real time polymerase chain reaction (real time PCR) technology. Westernblot and immunohistochemical assay respectively in asthmatic rats and control. [Result] Expression of AQP5 in lung decreased while MUC5AC increased more obviously in asthmatic rats than that of control and the difference was significant (P <0.01). There was significant negative relationship between the expression of AQP5 and MUC5AC in lung of asthmatic mice(r =0.901, P <0.01). [Conclusion] The decreased expression of AQP5 in lung may be the reason of increased expression of MUC5AC from the lung of asthmatic rats, which enhances the overproduction of airway mucus.
