摘    要:摘要] 目的: 研究注射环磷酰胺(CTX)大鼠同时应用促性腺激素释放激素激动剂(GnRHa)对注射环磷酰胺(CTX)大鼠的卵巢功能的影响.方法: 40只大鼠分为4组:空白对照组(10只),GnRHa组(10只),CTX组(10只), CTX + GnRHa组(10只)分别接受0.9%生理盐水、CTX、GnRHa和CTX + GnRHa注射。采用放射免疫分析法测定各组大鼠血清卵泡刺激素( FSH)和雌二醇(E2)浓度的变化.在停药时及停药后两周, 正常对照组各处死5只大鼠,其余三组分别处死4只大鼠. 观察卵巢重量、卵泡数量及卵泡直径的变化。结果:停药后两周,联合治疗组大鼠的血清雌二醇和FSH浓度分别为(43.39±4.40)pg/ml与(3.48±0.16)mIU/mL;与正常对照组比较,差异无统计学意义( P > 0.05);而CTX组大鼠在血清雌二醇和FSH浓度分别为(26.97±5.58)pg/ml与(3.92±0.35)mIU/mL ,与其余3组分别比较,差异均有统计学意义( P < 0.05)。在停药时和停药后两周处死大鼠的卵巢中,CTX组卵泡总数最少,各组间比较,差异均有统计学意义( P < 0.05) ;结论: 注射CTX同时应用GnRHa能够减少CTX对大鼠的卵巢功能的损伤。 关键词] 促性腺激素释放激素激动剂 环磷酰胺 卵巢 大鼠;

The affection of gonadotropin releasing hormone agonist used with CTX simultaneously on chemotherapy-induced ovarian damage in rats.
Abstract:Abstract]  AIM: To study the affection of gonadotropin-releasing hormone agonist ( GnRHa) used with CTX simultaneously against cyclophosphamide (CTX) induced ovarian damage in female rats. METHODS: Forty Wistar rats were divided into four groups, which received normal saline(NS),CTX, GnRHa and CTX + GnRHa respectively. Levels of serum follicil-stimulating hormone(FSH)and estradiol(E2) were measured by radio-immunity method. After stop of medication and two weeks later, half of the rats of the control group were killed and 4 rats were killed of other three groups, respectively to compare the weight of the ovaries and the number of the primordial follicles ,the growth follicles and the mature follicles. RESULTS: After two weeks of stop of medication, in the rats in the CTX + GnRHa group, the serum estradiol and FSH were (43.39±4.40)pg/ml and (3.48±0.16)mIU/mL respectively. Compared with the control group, there was no obvious difference between two groups ( P > 0.05).But in the rats in the CTX group, the serum estradiol and FSH were (26.97±5.58)pg/ml and (3.92±0.35)mIU/mL respectively. There was significant difference between the CTX group and other three groups.( P < 0.05) . After stop of medication and two weeks later , The follicle number of rats in CTX was least. And there was obvious difference between any two groups ( P <0.05). CONCLUSION: GnRHa used with CTX simultaneously can diminish cyclophosphamide (CTX) induced ovarian damage in female rats.
