鼻咽癌患者外周血白细胞及血清中EB病毒DNA的检测

摘    要:目的探讨鼻咽癌患者外周血白细胞及血清中EB病毒DNA的分布及其与鼻咽癌的关系.方法采用多聚酶链反应检测24例鼻咽癌患者(实验组)及10例头颈部其它部位肿瘤(对照组-1)和10例慢性鼻咽炎患者(对照组-2)外周血白细胞及血清中EB病毒的DNA.同时采用免疫酶法检测以上各组患者血清中EB病毒VCA-IgA抗体的效价.结果发现实验组中10例患者外周血白细胞中EB病毒DNA为阳性,阳性率为41.67%(10/24).对照组-1阳性率为10%(1/10),对照组-2患者外周血白细胞中EB病毒DNA均为阴性.实验组与对照组-2比较阳性率差异有显著性(P<0.05).在以上三组患者血清中EB病毒DNA均为阴性,三组间无差异.实验组VCA-IgA与对照组-1,对照组-2分别比较阳性率其差异均有显著性(P<0.01).结论有些鼻咽癌患者外周血白细胞中可检测出EB病毒DNA.血清中未能检测出EB病毒DNA.鼻咽癌患者外周血白细胞EB病毒DNA的存在与血清VCA-IgA的效价之间有一定的关系.

Detection of Epstein-Barr virus DNA in peripheral blood leukocytes and serum of patients with nasopharyngeal carcinoma

Abstract:Objective To study EB virus DNA distribution in peripheral blood leukocytes and serum in nasopharyngeal carcinoma patients(NPC). Methods Using polymerase chain reaction(PCR) method,24 patients withNPC, 10 patients with other head and neck carcinoma(control group 1) and 10 patients with chronic nasopharyngitis(control group 2) were tested in the presence of Epstein Barr virus DNA in peripheral blood leukocyte and serum. Meanwhile, serum EB virus VCA IgA antibodies were measured with enzyme immunoassay in all patients. Results 10 of 24 nasopharyngeal carcinoma patients(41 67%) and 1 of 10 control group 1(10%) were EB virus DNA positive in peripheral blood leukocytes. Control group 2 EB virus DNA was negative in peripheral blood leukocytes. The difference between NPC and control group 2 was statistically significant( P< 0.05). All of the patients' EB virus DNA were negative in serum. The difference between NPC and control group 1;2 was statistically significant in VCA IgA titer respectively( P< 0 01). Conclusion EB virus DNA is positive in peripheral blood leukocytes in some patients. But in serum the EB virus DNA copies are too weak to be detected. There must be some relationship between VCA IgA and EB virus DNA in peripheral blood leukocytes.
