小鼠急性四氯化碳肝损伤模型的建立及在保健食品筛选中的应用

摘    要:目的通过建立小鼠急性肝损伤模型,并将该模型应用于保健食品保护化学性肝损伤功能的筛选。方法 小鼠腹腔注射按1.0ml/100 g.b.w,一次分别给予0.063%、0.125%、0.25%、0.5%和1.0%四氯化碳(CCl4)溶液,24 h后取血,测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)含量变化;取肝脏常规病理制片。并利用以上实验所确立的小鼠急性肝损伤模型,测试样品的护肝功能。结果0.125%、0.25%、0.5%、1.0%CCl4各剂量组血清ALT与AST都比溶剂对照组高,差别有显著性意义(P<0.05)。CCl4各剂量组动物的肝脏病理变化以肝细胞气球样变及肝细胞坏死这两类型为主,其中0.125%、0.25%、0.5%、1.0%剂量组的肝脏病理损伤都比与溶剂对照组明显,差异有显著性意义(P<0.05)。用0.125%CCl4小鼠腹腔注射造模测试样品的结果显示,该受试样品具有一定护肝作用。结论建立了0.125%CCl4腹腔注射制造的小鼠急性肝损伤模型,可应用于保健食品护肝功能的筛选。

Abstract:Objective To establish the mice acute liver injury model induced by carbon tetrachloride and to apply in health food screening. Methods Carbon tetrachloride ( CCl4) 1.0 ml/100 g. b. w was given to Kunming mice by abdominal injection at the dose of 0.063% , 0. 125% , 0. 25% , 0.5% and 1.0% for one time. Mice ALT and AST in serum were analyzed 24 hours after administration. Mice liver was observed with routine pathology technique, to determine the liver protection function to mice. Results The serum level of ALT and AST increase significantly in 0.125%, 0.25% , 0.5% and 1.0% CCl4 groups compared with the control group (P < 0.05 ) . The pathological changes in 5 dosage groups were balloon degeneration and necrosis mainly, in which, 0.125% , 0.25% , 0.5% and 1.0% CCl4) groups were remarkably different with the control group ( P < 0.05) . The sample tested had certain liver protection function attacked with CCl4 at dose of 0.125% to mice. Conclusion 0.125% CCl4 mice acute liver injury model was established and could be applied to health food screening.
