牙釉质细胞型颅咽管瘤基因表达的基因芯片研究

摘    要:
目的建立牙釉质细胞型颅咽管瘤的基因表达谱,筛选差异表达基因,并探讨其与牙釉质细胞型颅咽管瘤的关系。方法采用cDNA微阵列分析牙釉质细胞型颅咽管瘤组织与正常垂体柄组织基因表达谱的变化,应用实时荧光定量PCR技术验证cDNA微阵列检测出差异基因的可靠性,分析牙釉质细胞型颅咽管瘤组织相关基因的差异表达。结果发现釉质细胞型颅咽管瘤与正常垂体组织之间差异性表达基因453条,其中前者表达下调258条,表达下调195条。结论牙釉质细胞型颅咽管瘤的发生可能涉及到细胞转录、细胞增殖、细胞凋亡等诸多方面,其相关基因表达的变化及其临床和病理学意义尚需进一步验证。

Abstract:
Objectives To establish the gene expression profile of adamantinomatous craniopharyngiomas and investigate the expression of the differential gene related to adamantinomatous craniopharyngiomas. Methods The difference in the gene expression profile between adamantinomatous craniopharyngiomas and normal tissue of hypophyseal stalk were analyzed by cDNA microarray which contained approximately 16 450 unigene that would be tested in the study. Real-time quantitative PCR technique was employed to validate the authenticity of cDNA microarray. Results Four hundred and fifty-three genes expressed in adamantinomatous cranio- pharyngiomas were different from the gene expressed in the normal hypophyseal stalk. Of these 453 genes, 195 genes were up- regulated and 258 were down-regulated in the adamantinomatous craniopharyngiomas compared to those in the normal hypophyseal stalk. Conclusions It is suggested that several genes are involved in the pathogenesis of adamantinomatous craniopharyngiomas. However, their relationship with the pathogenesis of adamantinomatous craniopharyngiomas should be studied further.
