刺五加关节腔注射治疗兔膝关节骨性关节炎的病理学观察

目的：观察关节软骨和滑膜的病理改变,探讨刺五加治疗原发性膝关节骨性关节炎的疗效及其作用机制。方法：将64只新西兰白兔随机分为4组：正常组、刺五加组、透明质酸钠组、模型组。正常组既不造模也不给药,其余用内侧半月板部分切除的方法造模。刺五加组关节腔注射刺五加注射液,透明质酸钠组关节腔注射透明质酸钠注射液。术后2周开始用药干预,直到处死前1周。分6周和10周两个时相点处死动物取材,制备软骨及滑膜石蜡切片后进行HE及AB-PAS染色,分别在光镜下进行病理评分。结果：滑膜Mankins评分,在两个时相点刺五加组均比模型组低（均P〈0.05）;与6周相比,刺五加组10周时评分较高（P〈0.05）。关节软骨OARSI评分,在两个时相点,刺五加组均比模型组低（均P〈0.05）;与6周相比,刺五加组10周评分无显著差异（P〉0.05）。结论：刺五加注射液可以减缓甚至阻断实验性兔膝关节骨性关节炎的病变进展。

Pathology Observation of Acanthopanax Senticosus on Knee Osteoarthritis of Rabbit

Objective：Through observing pathological changes of cartilage and synovium,explore curative effect and mechanism of Acanthopanax senticosus on primary knee osteoarthritis of rabbit.Methods：Sixty-four New Zealand white rabbits were randomly divided into normal group,Acanthopanax senticosus group,sodium hyaluronate group and model group.Normal group neither establish model nor gived drup,The remaining using the modeling method of resecting the part of medial meniscus.Acanthopanax senticosus group was injected Acanthopanax senticosus injection in the operative side knee joint weekly,sodium hyaluronate group was injected sodium hyaluronate in the same way.All regimens were treated at 2 weeks after operation until 1 week before the animal executed.At 6 weeks and 10 weeks both time points,all animals were killed,cartilage and synovial membrane paraffin section were made and stained with HE and AB-PAS,by light microscope,each section was evaluated and scored respectively.Result：Compared with model group,synovial membrane＇s Mankins score of Acanthopanax senticosus group is lower at both time points（P0.05）;Compared with 6 weeks time point,score of Acanthopanax senticosus group is higher at 10 week time point（P0.05）.Compared with model group,cartilage＇s OARSI score of Acanthopanax senticosus group is lower at both time points（P0.05）;There was insignificantly difference in Acanthopanax senticosus group between both the time points（P0.05）.Conclusion：Intraarticular injection of Acanthopanax senticosus can release even interrupt the development of lesion of knee osteoarthritis.
