表皮角质形成细胞分化中tPA分泌的研究

摘    要:目的探讨表皮角质形成细胞中,tPA分泌的变化及其对细胞分化的影响.方法利用免疫细胞化学、酶联免疫吸附测定等方法,检测高浓度Ca2 作用下,培养的小鼠分化表皮角质形成细胞表面及培养上清中tPA表达的变化.结果高Ca2 浓度下,KC分化标记物K10在培养24h即有较强的表达,与此同时,在培养上清中检测到大量分泌tPA;培养24h以后,随着培养时间的延长,KC培养上清中分泌tPA的含量呈现出降低趋势,KC表面tPA量则呈现出增加趋势,当有L-精氨酸存在时,上述两种趋势均明显减弱.结论小鼠表皮KC分化过程中存在tPA的分泌,分泌tPA能通过其赖氨酸位点进一步吸附于KC表面,进而促进KC的分化.

Abstract:Objective To explore the changes of plasminogen activator (tPA) secretion and its effects on epidermal keratinocyte (KC) differentiation. Methods The immunocytochemical method (ICC) and enzyme linked immunosorbent assay (ELISA) were used to determine the tPA amounts in culture supernatant as well as on cell surface of the differentiated mouse KCs cultured in high calcium medium. Results The KC differentiation marker keratin 10 was strongly expressed 24 h after KCs being cultured in high calcium medium, and a large amount of secreted tPA was detected in culture supernatant. A decrease in the amount of secreted tPA in culture supernatant and an increase in the amount of cell surface associated tPA were found after the KCs being cultured for 24 h, but the decrease and the increase could be inhibited by L arginine. Conclusion The tPA, secreted in the processes of KCs differentiation, can bind to the KCs surface through its lysine sites, and promote the tPA secreation.
