摘    要:目的 观察碱性成纤维细胞生长因子(bFGF)在体外培养的不同阶段小鼠晶状体上皮细胞(LECs)中的表达,及其对细胞周期的影响并探讨其机制。方法将昆明健康小鼠的晶状体前囊膜取出,做体外培养并传代。利用免疫组织化学法和Westernblot法检测bFGF在体外培养的不同阶段LECs中的表达情况,并利用流式细胞法检测相应阶段LECs的细胞周期情况。结果bFGF蛋白在LECs的原代、传第1、2代中明显表达,在传第3、4代中表达较弱。这与流式细胞法检测的细胞周期中有丝分裂相所占比例相符。结论bFGF参与LECs的增殖分化过程;并随着传代的递增,其促LECs增殖作用减弱,提示它与临床儿童后发性白内障发生率高有关。

Abstract:Objective To study the expression of basic fibroblast growth factor ( bFGF) in cultured lens epithelial cells (LECs) of mice. Methods Mouse lens anterior capsules with LECs were cultured in vitro,and the expression of bFGF in LECs was detected by immunohistochemistry and Western blots. Cell cycle of different stages was observed by flow cytometry. Results The protein of bFGF was strongly expressed in the first, second passage of LECs, and its expression was reduced in the third and fourth passage of LECs,showing a consistent proportion with mitosis in cell cycle. Conclusion bFGF might play an important role in the process of LECs proliferation, and bFGF is related to the high incidence of posterior capsular opacity (PCO) in children.
