原发性肝癌组织中三种凋亡相关基因的表达及意义

摘    要:
目的探讨原发性肝癌组织中三种凋亡相关基因:XIAP、Smac及Caspases-3的表达及意义。方法应用免疫组化方法检测67例原发性肝癌组织中XIAP、Smac及Caspases-3的表达情况。结果XIAP、Smac及Caspases-3在正常肝组织中的阳性率分别为(5.9±1.0)%、(90.1±0.8)%和(87.7±1.1)%,在肝癌组织中的阳性率分别为(89.7±0.7)%、(53.21±1.3)%和(49.8±0.9)%,三者在不同性质组织问差异均有统计学意义(t=7.78,5.37,6.01,P〈0.05);肝癌组织中XIAP与Caspases-3的表达呈负相关(r=-0.69,P〈0.05),Smac与Caspases-3的表达呈正相关(r=0.67,P〈0.05)。结论XIAP、Smac通过影响Caspases-3的表达拮抗细胞凋亡过程,参与原发性肝癌的发生、发展。

Abstract:
Objective To investigate the meaning of the expression of three proapoptosis genes.XIAP,Smac and caspases-3,in primary hepatic carcinoma.Method The expression of XIAP,Smac and Caspases-3 of 67 cases primary hepatic cancer was detected with immunohistochemistry.Result The posi-tive rate of XIAP,Smac and easpases-3 in tissues of hepatic carcinoma was(89.7±0.7)%,(53.21±1.3)%and(49.8±0.9)%respectively,while(5.9±1.0)%,(90.1±0.8)%and(87.7±1.1)%in normal hepatic tissues.And it had statistical significance(t=7.78,5.37,6.01,P<0.05).This expres-sion of caspases-3 negatively correlated with XIAP(r=-0.69,P<0.05)while positively with Smac(r =0.67,P<0.05).Conclusion XIAP and Smac play a role of anti-apoptosis by effecting the expression of easpases-3 and participate the genesis and development of the cancer.
