杜仲抗炎镇痛作用的实验研究

摘    要:目的研究杜仲水提物的镇痛和抗炎作用,为该药的临床研究提供基础。方法采用二甲苯所致的小鼠耳廓肿胀、蛋清所致大鼠足跖肿胀等炎症模型;热板法、乙酸诱导小鼠疼痛反应等疼痛模型,观察杜仲水提物的抗炎镇痛作用。实验动物每天灌胃一次,小鼠持续7d,大鼠持续15d。对照组给予蒸馏水,阿司匹林组给予阿司匹林(小鼠给予0.25g/kg、大鼠0.18g/kg),盐酸曲马朵组给予盐酸曲马朵0.15g/kg,高、中、低剂量组分别给予杜仲水提物(小鼠给予163.80、81.90、40.95g生药/kg;大鼠给予114.66、57.33、28.67g生药/kg)。分别在末次给药后进行镇痛和抗炎作用的观察。结果杜仲能显著的延长醋酸所致小鼠的疼痛扭体首次出现的时间,减少扭体次数和提高扭体抑制率和镇痛百分率,延长热板致痛小鼠的痛阈值;高剂量组的作用与阿司匹林相当,但镇痛效果不如盐酸曲马朵,呈现良好的剂量依赖关系。同时,在一定程度上杜仲能明显的抑制蛋清所致大鼠的足肿胀和二甲苯致小鼠耳廓肿胀,减轻小鼠因炎性物质刺激导致的炎性渗出,作用依然呈现良好的量效趋势,高剂量组的作用与阿司匹林相当。结论杜仲具有显著的抗炎作用和镇痛作用,这些药理作用为其临床治疗疼痛和炎症性疾病提供了实验依据。

ANTI-INFLAMMATORY AND ANALGESIC EFFECTS OF WATER EXTRACT FROM EUCOMMIA ULMOIDE OLIVER IN MICE

Abstract:Objective To explore analgesia and anti - inflammatory effects of the water extract from Eucommia ulmoides on mice. Methods Different animal models were established to observe the analgesic and anti - inflammatory effects of Eucommia ulmoides. The animal models included dimethylbenzene - induced acute ear swelling in mice, ovi albumin - induced toe swelling in rats;pain experiments induced by hot plate and glacial acetic acid in mice 3 test groups were given with the water extract ( mice :163.80,81.90,40.95 g / kg, rats: 114.66,57.33,28.67g/kg ), respectively by gavage once a day for 7 days (mice) or 15 days (rats), aspirin group were given with the aspirin(mice: 0. 25g/kg,rats.- 0. 18g/kg) ,ultram group were given with the ultram 0. 15g/kg,in the same manner, normal control group were given with the same volume of water. After the seventh day of giving drug, the mice were experientized. Results Euecommia ulmoides could significantly relieve'the pain caused by glacial acetic acid and heat stimuli; It also showed obvious inhibitory effects on ovi albumin - induced toe edema in rats and dimethyl- benzene- induced ear swelling in mice. At the same time, the results showed a clear dose - dependent relation in the mice of test groups, the effect of the high-dose group was the best, which of the low-dose group was the worst. Conclusion The ethanol extract from Eueommia ulmoides show markedly analgesic and anti - inflammatory effects in the study, which provides the basis for the clinical utilization in the treatment of ache and phlogosis diseases.
