氪激光诱导大鼠脉络膜新生血管基因芯片研究

摘    要:目的运用基因芯片技术研究氪激光诱导大鼠脉络膜新生血管的差异表达基因.为脉络膜新生血管的治疗提供实验依据。方法氪激光光凝视网膜后7d,分剐提取实验组和对照组脉络膜组织的总RNA,将纯化后的mRNA进行反转录,制备杂交探针,应用含有5705条Oligo DNA的表达谱芯片对实验组和对照组脉络膜组织进行差异表达基因检测。结果氪激光诱导脉络膜新生血管组织的差异表达基因共有427条,其中上调基因为189条,下调基因为238条。上调最高的基因是MMP12基因。在差异表达基因中有代谢、细胞粘附、细胞运动、发育、运输、信号传导、分化等基因。结论氪激光诱导脉络膜新生血管的发生发展涉及多基因改变。[眼科新进展2005;25(5):408—410]

Abstract:Objective To explore different gene expression of choroidal neovascularization with gene chip technology and to provide experimental evidence for therapy of choroidal neovascularization.Methods On the 7th day after krypton laser photocoagulation, total RNAs of experimental group and control group were extracted from choroid tissue.The mRNA was purified and then was reversely transcribed to cDNA for hybridization probes. The differential expressed genes in choroidal tissue of experimental group and control group were assayed with gene chips which contained 5 705 rat genes.Results There were 427 differential expressed genes in which 189 genes were upregulated and 238 genes downregulated.MMP12 was the most tallest gene in upregulated genes.The differential expressed genes were involved in metablolism,cell adhesion,cell motility development,transportation,signal and differentiation genes.Conclusion The genesis of choroidal neovascularization induced by krypton laser photocoagulation was involved alteration in many genes.
