摘    要:目的:探讨内脂素(visfatin)过表达对多囊卵巢综合征(PCOS)模型大鼠性激素水平变化及代谢方面的影响。方法:采用42日龄SD清洁雌性大鼠注射脱氢表雄酮(DHEA)和胰岛素诱导建立PCOS模型(n=20),并以等量油剂代替DHEA为对照组(n=20),大鼠visfatin基因真核表达质粒和空质粒分别转染PCOS组和对照组,观察卵巢相对重量及光镜(HE染色)形态学改变,ELISA方法测定血浆visfatin水平,放射免疫分析法测定空腹血浆胰岛素(FIns)含量及T、LH、FSH,并计算胰岛素抵抗指数(HOMA-IR)指数变化。结果:visfatin质粒注射72 h后较注射前,血浆visfatin水平升高(P<0.05)。卵巢重量显著降低(P<0.05),卵巢见多个黄体及不同发育阶段的卵泡,血清T、LH、空腹胰岛素及胰岛素抵抗指数(HOMA-IR)显著降低(P<0.05)。结论:visfatin质粒转染在PCOS模型大鼠中起部分调节作用,并与胰岛素抵抗有关。

Abstract:Objective:To explore the effect of overexpression of visfatin on changes of sex hormones levels and metabolism of rat models of polycystic ovarian syndrome(PCOS).Methods:Twenty SD female rats aged 42 days old were treated with injection of dehydroepiandrosterone(DHEA) and insulin to construct PCOS model,and other twenty SD female rats aged 42 days old were treated with oil of the same dose and insulin to construct PCOS model,and then they were designed as control group,the rats in PCOS group and control group were transferred by eukaryotic expression plasmid of rat visfatin gene and blank plasmid,respectively,the relative weight of ovary and morphological changes under light microscope were observed,ELISA was used to detect plasma visfatin level,radioimmunoassay was used to measure the contents of fasting serum insulin,testosterone,luteinizing hormone,and follicle-stimulating hormone(FSH);the change of insulin resistance index was calculated.Results:Compared with before injection of visfatin plasmid,plasma visfatin level increased significantly at 72 hours after injection,there was statistically significant difference(P<0.05).Ovarian weight decreased significantly(P<0.05),many corpus luteum and follicles at different developmental periods could be found in ovary.The concentrations of serum testosterone,luteinizing hormone,fasting serum insulin,and insulin resistance index decreased significantly,there was statistically significant difference(P<0.05).Conclusion:Visfatin plasmid transfection plays the part of regulatory role in rat models of PCOS,and the effect is correlated with insulin resistance.
