福辛普利、氯沙坦对自发性高血压大鼠脂肪组织脂联素的影响

摘    要:目的:探讨福辛普利和氯沙坦对自发性高血压(SHR)大鼠脂肪组织脂联素的影响。方法:22周龄SHR大鼠随机被分成四组.分别予福辛普利、氯沙坦、福辛普利加氯沙坦、安慰剂喂养。同系魏凯氏(WKY)大鼠作对照组。8周后取尿检测大鼠尿微量蛋白(Upro)和n-乙酰β-d-氨基葡萄糖苷酶(NAG)含量,取脂肪组织用免疫组化法及逆转录聚合酶链式反应(RT-PCR)检测脂联素的mRNA的表达。结果:SHR大鼠脂肪组织脂联素表达较WKY大鼠降低;Upro和NAG酶水平升高(P均〈0.01);福辛普利组和氯沙坦组与SHR组比较血压降低(P〈0.01);Upro和NAG酶水平降低(P〈0.01),脂联素表达上调(P〈0.01)。结论:(1)SHR大鼠Upro和NAG酶水平升高.脂肪组织脂联素表达下调;(2)福辛普利和氯沙坦可以提高SHR大鼠脂肪组织脂联素表达,降低Upro和NAG酶水平。

Abstract:Objective: To observe the impacts of fosinopril and losartan on adiponectin expression in adipose tissue of vascular walls of spontaneously hypertensive rats (SHR). Methods: Twenty two weeks old male SHR were randomly divided into four groups: Model (SHR) group, fosinopril (Fos) group, losartan (Los) group and fosinopril+losar-tan (Fos+Los) group; Five 22-week old male Wistar-Kyoto rats (WKY) were regard as the control group. After 8 weeks, 24 h urinary protein (Upro), urinary N-acetyl-β-d-glucosaminidase (NAGase) were detected; The mRNA ex- pression of adiponectin was detected by RT--PCR; the protein expression of adiponectin was tested by immunohisto chemistry. Results: As compared to WKY group, 24h urinary protein content and urinary NAGase were increased, adiponectin expression in adipose tissue of vascular walls was down-regulated in SHR groups (P〈0. 01 all) ; as com- pared to SHR group, blood pressure was lowered, 24h urinary protein content and urinary NAGase decreased, adi- ponectin expression was up-regulated in Fos, Los and Fos+ Los group (P〈0. 01 all). Conclusion: Urinary protein content and urinary NAGase increased, adiponectin expression in adipose tissue of vascular wails is down-regulated in SHR group; Fosinopril and/or losartan can decrease urinary protein content and urinary NAGase, up--regulate adi-ponectin expression in adipose tissue of vascular walls of SHR.
