牛磺胆酸钠致大鼠急性出血坏死性胰腺炎胰腺组织的光镜和电镜观察

摘    要:作者以5%的牛磺胆酸钠(Na-Tc)注入大鼠胆胰导管内引起急性出血坏死性胰腺炎。光镜和电镜观察结果显示:注入Na-Tc后10分钟到1小时,胰腺小叶出现细胞膜性结构溶解的灶性坏死,血管少许破坏和轻度出血;注入Na-Tc后3到 6小时,胰腺多个小叶成片坏死,血管溶解和重度出血。坏死前的胰腺细胞内出现大量含有酶原颗粒等细胞器的自噬泡。

OBSERVATION OF LIGHT AND ELECTRON MICROSCOPIC STRUCTURE OF PANCREAS IN ACUTE HAEMORRHAGIC NECROTIZING PANCREATITIS INDUCED BY SODIUM TAUROCHOLATE IN RATS

Abstract:Acute haemorrhagic necrotizing pan-creatitis was caused by injecting 5% sodiumtaurocholate (Na-Tc) directly into thecommon biliopancreatic duct in rats.In thecontrol group 0.9% NaCl was used. Lightmicroscopy on the pancreatic pathologictissue and electron microscopy on the pa-thologic changes of ultrastructure of thepancreatic acinar cells were carried outcontinuously. In the experimental group, light andelectron microscopical observation on pan-creatic lobule showed focal necrosis,dissolu-tion of membrane structure except that ofzymogen granules, mild damage of smallblood vessels and haemorrhage at 10, 30 and60 min after injection. A lot of pancreaticlobuli were characterized by extensivenecrosis, dissolved blood vessels wall andsevere haemorrhage, at 3h and 6h afterinjection. In the pancreatic acinar cells wasobserved the presence of a quantity of auto-phagic vacuoles which contained the resi-dual structure of the rough endoplasmicreticulum, mitochondria, zymogen granulesand altered zymogen granules at 3,6 h afterinjection. The results demonstrated that at firstthe damage was due to the detergent effectof Na-Tc leading to local cellular necrosisand vascular lesions in the pancreatic lobuliand afterward the damage was induced by"autodigestion" of pancreatic parenchyma.Appearance of autophagic vacuoles inthe pancreatic acinar cells suggested thatthe intracellular digestive zymogens be ac-tivated and the activated zymogens initiateautodigestion of the pancreas.
