何首乌、三七提取物对非酒精性脂肪性肝炎大鼠肝组织PPAR仅、PGC-1α水平的影响

摘    要:目的:探讨何首乌、三七提取物预防大鼠非酒精性脂肪性肝炎及其肝组织PPARⅡ、PGC-1α表达的影响。方法:32只雄性SD大鼠,随机分为4组,每组8只。除正常对照组外,高脂饲料喂养10周建立大鼠非酒精性脂肪性肝炎模型。于造模即日起,正常对照组、模型组每天分别给予10mL/kg饮用水,中药组给予何首乌、三七提取物,易善复组给予易善复灌胃。给药10周后,处死所有大鼠,检测血清LDL、HDL、TG,ALT、AST、GGT1肝组织TG和PPARα、PGC-1αmRNA水平。结果:中药组血清LDL含量较模型组下降(P〈0.05);中药组血清HDL、TG,ALT、AST、GGT和肝组织TG含量较模型组显著下降(P〈0.01),肝组织PPARα、PGC-1αmRNA水平较模型组显著增高(P〈0.01)。结论:何首乌、三七提取物可降低NASH大鼠血清、肝脏脂质合成与聚积,减少炎症反应,从而有效防治NASH,具有良好的临床应用前景。

Abstract:Objective To investigate the effect of the extracts of Heshouwu and Sanqi (Polygonum multiflorum Thunb.and Panax no toginseng)on the expression of peroxisome proliferators-activated receptor α (PPAR α) and PPAR gamma coactivator-1(PGC-1α) in non-alcoholic steatohepatitis (NASH) in rats. Methods The 32 male SD rats were randomized into 4 groups with 8 in each. Rat model of non-alcoholic steatohepatitis was established with fatty diet of 10 weeks except the normal control group. At the same time, the black group and model group were treated with the drinking water (10 mL/kg,daily) . The herbal group and Yishanfu group were treated with the extractive of Heshouwu and Sanqi, and Yishanfu respectively. All the rats were executed after the therapy of 10 weeks. The LDL, HDL, TG, ALT, AST and GGT in the blood serum, TG, PPAR α and PGC-1α mRNA in the liver were detected. Results Compared to the model group, the LDL in the blood serum in the herbal group decreased (P 〉 0.05).HDL,TG, ALT, AST and GGT in the blood serum, TG in the liver in the herbal group showed significant difference (P 〉 0.01), while significantly increased (P 〈 0.01) in the PPARαand PGC-1α mRNA in the liver. Conclusion The extractive of Heshouwu and Sanqi can degrade lipid synthesis and accumulation in blood serum and liver of NASH rats, and decrease inflammatory reaction at the same time. The extractive of Heshouwu and Sanqi can prevent and cure NASH, and could be used in clinic medicine in the future.
