摘    要:目的观察不同波长的紫外线照射对小鼠皮肤(胶原纤维和弹性纤维),对组织学及生物化学的影响,以探讨紫外线损伤皮肤的机制。方法用不同波长的紫外线照射小鼠皮肤12周后,测定各组小鼠皮肤丙二醛(malondialdehyde,MDA)含量。用V-G、Weigert染色方法分别显示胶原纤维和弹性纤维。用体视学测量和分析各组小鼠背部皮肤胶原纤维和弹性纤维的体密度、表面积密度和交点密度的变化。结果长波长紫外线(ultraviolet A,UVA)、中波长紫外线(ultrariolet B,UVB)照射组胶原纤维体密度、交点密度及表面积密度均低于对照组(P<0.01)。UVA照射组与UVB照射组胶原纤维变化差异无统计学意义(P>0.05)。UVA、UVB照射组弹性纤维体密度、交点密度及表面积密度、皮肤MDA含量均高于对照组(P<0.01)。结论UVA与UVB照射可使小鼠皮肤MDA、胶原纤维和弹性纤维发生改变,但两者短期内对小鼠皮肤MDA、胶原纤维损害的差异性不明显。

Abstract:Objective To investigate the histological and biochemical changes of mice skin and possible mechanism irradiated by ultraviolet of different spectrum. Method Using special stain technique, and biochemical and stereological methods, the changes of mice skin were observed after irradiated with ultraviolet A or ultraviolet B or un-ultraviolet for 12 weeks. Results The most significant findings were obviously lower level of the volume density and surface area density and intersection density of collagen fibers in ultraviolet A (UVA) irradited group and ultraviolet B (UVB) irradited group (P<0.01), and significantly higher level of the volume density and surface area density and intersection density of elastic fibers in ultraviolet A irradited and ultraviolet B irradited group (P<0.01). The level of MDA in control group was lower than that in UVA group and UVB group (P<0.01). Conclusions Ultraviolet irradition could damage collagen fibers and elastic fibers and enhance the level of MDA in mice skin, but the difference of collagen fibers between UVA and UVB is not significiant.
