银杏叶提取物对兔外周血干细胞动员的影响

摘    要:目的:探讨银杏叶提取物对兔外周血干细胞的动员作用。方法:30只2628周龄新西兰大白兔,体重为2.528周龄新西兰大白兔,体重为2.53.0kg,随机分三组:银杏叶提取物组(Egb)、G-CSF组、生理盐水组,给药方法为腹腔注射7d,1次/d;给药后23.0kg,随机分三组:银杏叶提取物组(Egb)、G-CSF组、生理盐水组,给药方法为腹腔注射7d,1次/d;给药后28d,采用外周血WBC和MNC计数、造血干细胞体外培养、免疫细胞化学等检测各组外周血WBC、MNC、CD34+细胞、CD49d阳性细胞、CFU-GM、CFU-MK、CFU-E的产率及兔骨髓基质细胞中VCAM-1阳性细胞百分率。结果:Egb组给药第7天外周血WBC、MNC数量达到高峰,分别为给药前的3倍和3.5倍;Egb组CD49d阳性细胞、外周血CD34+及骨髓基质细胞的VCAM-1阳性细胞百分率分别为(9.37±3.01)%、(0.63±0.37)%和(48.13±9.84)%,均高于生理盐水组;其中CFU-GM、CFU-MK和CFU-E产率分别为(12.94±3.03)%、(11.49±2.93)%和(24.52±8.62)%,均高于生理盐水组。结论:银杏叶提取物(Egb)对兔骨髓造血细胞有一定的增生作用,同时对外周血干细胞有一定的动员作用,这可能与其上调兔骨髓基质细胞和外周血粘附分子的表达有关。

The effects of peripheral blood stem cell mobilization on rabbits with ginkgo biloba extract

Abstract:Objective :To study Ginkgo biloba extract (Egb) for mobilizing peripheral blood hematopoiet-ic stem cells and the effects of Egb on the expression of adherent molecule of peripheral blood hematopoietic stem cells and bone marrow cells in rats .Methods :Thirty rabbits were divided into three groups at random (n= 10 each):rabbits were intraperitoneally injected with Group Egb and rhG-CSF (Group G-CSF) respectively from 1 day to 7 day ,normal control with equivalence saline .The effects of the numbers of MNC and WBC the percentage of CD34+ cell and CD49d positive cell and the yields of CFU-GM、CFU-MK、CFU-E in peripheral and the percentage of VCAM-1 positive cells in bone marrow stromal cells of the mice were observed by means of WBC and MNC counts , flow cytometry ,hematopoietic cell culture in vitro and immunocytochemistry .Results :7 days after be treated of Egb group ,the numbers of WBC and MNC reached the maximum ,which were 3 and 3 .5 times that of the NS group re-spectively ;the perce-ntage of CD34+ cells and CD49d positive cells in peripheral blood and VCAM-1 positive cells in bone marrow stromal cells in Egb group was (0 .63 ± 0 .37)% 、(9 .37 ± 3 .01)% and (48 .13 ±9 .84)% respectively , were markedly higher than in NS group ;the yields of CFU-GM 、CFU-MK and CFU-E in group of Egb was (12 .94 ± 3 .03)% 、(11 .49 ± 2 .93)% and (24 .52 ± 8 .62)% respectively ,were markedly higher than those in NS group . Conclusion :Egb could mobilize hematopoietic stem cells into peripheral blood ,which role may be related to increase adhesion molecule expression in peripheral blood cells and bone marrow stromal cell in rabbits .
