摘    要:目的建立测定血清中奥氮平浓度的反相高效液相色谱(RP-HPLC)法。方法血清碱化后用乙醚提取,采用RP-HPLC法检测,色谱柱为Hypersil ODS柱(150mm×4.6mm,2.5μm),流动相为磷酸盐缓冲液(pH=6.86,内含0.5%三乙胺)-甲醇(40:60,V/V),流速为1.0mL/min,检测波长为270nm,柱温25℃。结果血清中奥氮平质量浓度线性范围为10.16~203.00μg/L(r=0.9998),最低检测质量浓度为1.22μg/L,相对回收率为99.4%~100.6%,日内RSD为2.00%~2.29%(n=5),日内RSD为2.05%~4.28%(n=5)。结论该方法简便、准确,重现性好,灵敏度高,适用于奥氮平血药浓度的监测。

Abstract:Objective To develop a RP- HPLC method for the determination of olanzapine concentration in serum. Methods The alkalified serum samples were extracted with diethyl ether, the separation was made by using a reversed-phase Hypersil ODS column with isocratic mobile phase consisting of phosphat buffer (pH = 6.86, containing 0, 5% triethylamine) - methanol (60 : 40, V/V) ;the flow rate was 1.0 mL/min and the detective wavelength was 270 nm. The temperature of the column was 25℃. Results The linear range was 10. 16- 203.00μg/L (r=0.999 8), the lowest detectable concentration in serum was 1.22μg/L. The recovery rates were 99.39%- 100.59%.The intra-day RSD was 2.00% - 2.29%. The inter- day RSD was 2.05% -4.28%. Conclusion The established method is sensitive, precise, reproducible, and suitable for the TDM of olanzapine.
