摘    要:
目的观察小檗碱对人胃癌细胞BGC-823凋亡的诱导作用,以探讨具抗癌作用机理.方法采用体外细胞培养技术,以lO~40 mg/L小檗碱作用于培养的BGC-823细胞48 h后,进行光镜、琼脂糖凝胶电泳、流式细胞仪和TUNEL法的观察和分析.结果小檗碱作用后,光镜、TUNEL观察到细胞呈现凋亡特征,琼脂糖凝胶电泳显示细胞DNA呈梯状降解,流式细胞仪分析出现典型亚二倍体峰.结论小檗碱可诱导人胃癌细胞BGC-823细胞凋亡,这可能为小檗碱抗癌的机理之一.

Abstract:
Objective: To investigate the mechanism of the antitumor effect of berberine by observing the apoptosis of human gastric carcinoma BGC- 823 cells induced by berberine. Methods:By technique of cell culture in vitro, BGC -823 cells were treated by berberine in 10 ~ 40 mg/L concentration for 48 hours, then observed and analyzed by light microscopy, agarose gel electrophoresis, flow cytometry and Tunnel. Results:The cells showed characters of apoptosis observed by light microscopy and Tunnel. agarose gel elctrophoresis of DNA showed change of DNA ladder'pattern. A typical subdiploid peak before G0/Gl phase was observed by flow cytonetry. Conclusions:The results confirm that the berberine could induce the apoptosis of BGC -823 cells, which may be one of the mechanisms of bcrberine's antitumor effects.
