三七总皂苷对大鼠慢性高眼压视网膜损伤的保护作用

摘    要:目的探讨三七总皂苷对实验性高眼压导致的大鼠视网膜神经节细胞层(RGCL)神经元损伤有无保护作用。方法健康SD大鼠42只,其中6只为正常对照组,6只为正常用药组(三七总皂苷200mg/kg),其余30只用Akira法,烙闭大鼠右眼上巩膜静脉,制作大鼠持续性高眼压模型后随机分为生理盐水组和量效关系组,分别予tPNS50mg/kg(A组),100mg/kg(B组),150mg/kg(C组)和200mg/kg(D组)治疗1个月。处死大鼠剥取视网膜做全层铺片,进行RGCL神经元计数分析。结果正常对照组大鼠双眼RGCL神经元计数比较差异无显著性意义(P>0.05),正常对照组与正常用药组比较差异无显著性(P>0.05),正常对照组与各浓度tPNS治疗组实验眼RGCL计数差异均有显著性(P<0.05),各浓度tPNS治疗组实验眼与其自身对照眼RGCL计数差异均有显著性(P<0.05),生理盐水组与C组实验眼RGCL计数差异有显著性(P<0.05)。结论单独使用tPNS对正常大鼠RGCL神经元计数无影响,tPNS对持续性高眼压大鼠RGCL神经元损伤有部分保护作用。RGCL中的大神经元对高眼压更敏感,与青光眼RGC丧失的模式相似。

Abstract:OBJECTIVE To investigate if tPNS has a neuroprotective effect against chronic ocular hypertension induced neuron death in RGCL.METHODS Forty-two healthy SD rats were randomly divided into seven groups.Six were in normal control group,while another 6 were in normal treated group(tPNS200 mg/kg)and the others were cauterized three episcleral vessels and were divided into experimental groups:group A(50 mg/kg),group B(100 mg/kg),group C(150 mg/kg),group D(200 mg/kg)and group treated by normal saline.Rats were sacrificed on schedule.Flat preparation of whole retinas were stained with 0.5 %cresyl violet and neuron counting in RGCL from both eyes of each rat were performed to evaluate neuron loss.RESULTS Results There was no significant difference between the right eye and the left eye of normal control group(P>0.05).There was no significant difference between normal control group and normal treated group either in the right eye or in the left eye(P>0.05).There was no significant difference between normal control group and experimental groups(P>0.05).There was significant difference between group C and normal saline group(P<0.05).CONCLUSIONS When administered alone to normal rats,tPNS has no effect on neuron counting of RGCL,but it has partial protective effect on chronic ocular hypertension induced neuron death in RGCL in the rat.
