| 摘 要： | 本文报道了用人载脂蛋白A-1(apo A-1)的cDNA探针从人基因库中筛选到apo A-1全基因:将2.2kb的apo A-1全基因连接到1.9kb鼠MT-1基因启动子下游,组装成人apo A-1基因表达质粒pLY1。质粒pLY1与质粒pSV2-neo共转化中国仓鼠卵巢(CHO)细胞,经G418筛选,单细胞克隆培养,经酶联免疫测定,结果表明,人apo A-1可在CHO细胞中得到表达。 |

| Abstract: | The human apolipoprotein A-1 (apo A-1) gene was isolated from a genomic library by screening with cDNA probe. The expression plasmid including apo A-1 gene (2.2 kb) plus promoter (1.9kb) fragment of rat metallothionein 1 gene was transfected into Chinese Hamster Ovary (CHO)cells. Transfected cells were selected against neomy-cin resistance and screened for the ability to produce human apo A-1 by enzyme-linked immu nosorbent assay. Isolation of a clonal cell line resulted in the expression and secretion of human apo A-1. |
