结肠癌组织中p53和cyclinD1基因的表达检测及其临床意义

摘    要:
目的研究结肠癌、腺瘤和正常肠粘膜组织中p53和cyclinD1基因的表达及其临床意义。方法取85例结肠癌、18例结肠腺瘤和9例癌旁正常结肠粘膜组织分别制成72点和104点两块组织芯片,采用免疫组织化学法检测结肠组织芯片中p53和cyclinD1的表达。结果p53、cyclinD1在结肠癌、结肠腺瘤和正常结肠粘膜组织中的表达差异显著(P=0.003,P=0.000),p53、cycllnD1在结肠癌中的表达高于结肠腺瘤和正常结肠粘膜组织,不同组织学分化程度的结肠癌中p53的表达差异显著(P=0.005)。cyclinD1的表达与结肠癌临床分期有关(P=0.038),与其他临床病理资料无关。结论p53畸变可能是结肠癌发生的较晚期事件,cyclinD1高表达可与大肠癌发生有关.cvclinD1高表达的肿瘤更易浸润和发展。

Abstract:
Objective To study on the expressions and clinical significance of p53 and cyclinD1 in colon cancer,adenomatous polyps and normal para-cancer colon mucosa.Methods Eighty five cases of colon cancer,18 cases of adenomas and 9 cases of para-mucosa were accumulated and made into two tissue microarrays which containing 72 dost and 104 dots respectively.Expressions of p53 and cyclinD1 proteins were detected by immunohistocheical staining to these microarrays.Results Significant differences were found among colon cancer,adenomas and benign para-cancer mucosa according to the expressions of p53 and cyclinD1(P=0.003,P=0.000).Expressions of p53 and cyclinD1 in cancer were highter than that in adenomatous polyps and normal para-cancer colon mucosa.A significant difference of expressions of p53 in different histological grades(P=0.005) was found in colon cancer.There was no correlation of expression of cyclinD1 with clinicopathological data but clinical stages(P=0.038) in colon cancer was found.Conclusions Overexpression of p53 may be a late event involving in the development of colon cancer an high expressions of cyclinD1 may correlate with the tumorgenesis in colon and may facilitate invasion and progression.
