痕量氟化物快速测定方法建立

摘    要:目的 建立准确、快速、经济、实用的微量、痕量氟化物检测方法。方法 采用酶标仪-氟试剂分光光度法,测定生物、食品、环境等样品或其消化处理液中的氟化物含量,并以3水平4因素正交设计对该法的试剂浓度、比例、用量进行多因素分析,以优化最佳实验条件;以重现性、加标回收率和检出限评价方法的精密度、准确度及灵敏度。结果 最佳实验条件为氟试剂与硝酸镧比例为1:1,稳定剂为丙酮-乙酰丙酮,缓冲液3.5 mol/L;利用该方法可精确测定微量氟(氟含量>0.06μg/mL),痕量氟(氟含量≤0.06μg/mL);r为0.998 6~0.999 1,检出限为0.000 9μg,检测精度可达0.001μg,回收率为97.6%~100.1%,变异系数<2.65%。结论 将酶标仪引入氟化物的检测,分析速度快,检出限低,且具有较高的精密度、准确度。

Abstract:Objective To establish amore economical,rapid,practical,and accurate method for detection of trace fluoride in biological,food and environmental samples.Methods The trace fluorideion in biologic,food and environmental samples or the digestive liquid wasm easured with microplate reader fluoride reagent spectrophotometry.The optmium expermiental conditions on reagent concentration,ratio and amount were decided with a 3 level of 4 factor orthogonal expermient design.The precision,accuracy sensibility,reproducibility,expermient linearity,and recovery rate were also determ ined by adding standard material into the samples.Results The new method could accurately measure the fluoride at the concentration of less than 0.06 g/mL.The correlation coefficient was 0.998 6 0.999 1.The lmiitation of the detection was 0.000 9 g.The testing accuracy was 0.001 g.The recovery rate was between 97.6% 100.12% and the coefficient variation was 2.65%.Conclusion The rapid,practical,economic,and simple method with a micro plate reader can be used to test trace fluoride in biological,food and environmental samples.The method is more accurate,precise,sensitive,and less tmie-consuming.
