创伤性脑损伤后人脑组织中诱导性一氧化氮合酶蛋白水平的研究

摘    要:
目的:探讨创伤性脑损伤(Traumatic Brain Injury,TBI)后诱导型一氧化氮合酶(induced nitric oxide syn-thase,iNOS)在人脑组织中的表达变化及意义。方法:把38例脑损伤患者以及对照组患者分为7组(包括对照组,伤后0 ̄6h组、6 ̄12h组、12 ̄24h组、24 ̄48h组、48 ̄72h组,72h以上组)。用尼氏法染色观察脑损伤后神经细胞病理变化过程,免疫组织化学染色方法观察iNOS阳性细胞的表达变化。结果:尼氏法染色发现损伤区神经细胞减少;免疫组化检测发现伤后不同时相iNOS阳性细胞表达增加,伤后12 ̄24h达到高峰。结论:脑损伤后神经元的数量减少,损伤区及周围皮质出现iNOS阳性神经元,iNOS阳性细胞的表达与伤后时程有关。

Abstract:
Objective:To investigate the meaning and time-course change of inducible nitric oxide synthase(iNOS) expression in the damaged area after Traumatic humans Brain Injury.Methods: 38 Patients whose brains were injured and 4 normal persons are divided into 7 groups(including 0-6 hour group,6-12 hour group,12-24 hour group,24-36 hour group,and 36-72 hour group after the injury. The pathological changes of nerve cells in the damaged area were observed with Nissle. The expression of iNOS positive cells was observed by immunohistochemistry of different phases after contusion and laceration of the brain.Results: The neurons were decreased in the damaged area after injury observed with Nissle; iNOS positive cells observed by immunohistochemistry increased postinjury after contusion and laceration of the brain,and reached its peak during 12-24 hours. Conclusions: After Traumatic Brain Injury, the neurons were decreased in damaged area .There are positive neurons in the injured cortex ,but no positive neuronsin in the normal cortex.After Traumatic Brain Injury,the number of iNOS positive cells were associated with temporal course of postinjury.
