摘    要:目的:建立高效液相色谱串联质谱法(HPLC-MS/MS)测定人血浆中利奈唑胺的血药浓度,为临床合理用药提供参考。方法以替米沙坦为内标,血浆样品经乙腈沉淀蛋白进行提取分离,以含0.1%甲酸的1 mmol/L乙酸铵溶液和乙腈为流动相,梯度洗脱,采用m/z 338.1→m/z 296.3(利奈唑胺)和m/z 515.3→m/z 276.3(替米沙坦,内标)离子对进行扫描。结果利奈唑胺在0.23~46μg/ml线性范围内关系良好,r=0.9993,最低定量限为46 ng/ml,提取回收率为87%~109%,日内、日间精密度的相对标准偏差均在2.5%~8.6%范围内。结论建立的LC-MS/MS方法专属性强,灵敏度高,操作简单,适于临床利奈唑胺血药浓度监测。

Abstract:Objective To determine the concentration of linezolid in human plasma by high performance liquid chromatagra?phy-tandem mass spectrometry(HPLC-MS/MS)so as to provide evidence for the rational administration in clinic. Methods The plas?ma was extracted by acetonitrile with telmisartan as internal standard(IS). The mobile phase was 1 mmol/L ammonium acetate with 0.1%formic acid-acetonitrile with gradient elution. Quantitative analysis of[M+H]+ions was m/z 338.1→m/z 296.3(linezolid)and m/z 515.3→m/z 276.3(telmisartan,the IS). Results Excellent linear relationship was obtained from 0.23 to 46μg/ml with correlation co?efficient value of 0.9993. The lowest limit of quantitation was 46 ng/ml,the extraction recovery was from 87%to 109%. The intra-and inter- day precisions(relative standard deviations)ranged from 2.5% to 8.6%. Conclusion The method is specific,sensitive and simple and can be used to determine the concentration of linezolid in clinic.
