胰腺癌患者手术标本及胰液脱落细胞端粒酶活性的检测

目的 研究胰腺良，恶性患者手术标本，经ERCP所获胰液脱落细胞的端粒酶活性的表达，探索胰腺癌诊断的新方法。方法 采用TRAP-ELISA的方法检测胰腺癌细胞株，胰腺良恶性疾病组织物，胰液细胞的端粒酶活性。结果 6株胰腺癌细胞株均检测到了端粒酶活性，胰腺癌手术标本端粒酶的阳性率为63%（5/8），而4例正常胰腺组织和2例胰腺良性疾病患者的胰腺标本未检测到端粒酶活性，胰腺癌患者的胰液细胞中，有58%（11/19）检测到有端粒酶活性的表达，而胰腺良性疾病也有高达62.5%(5/8)的阳笥率，两者无差别。结论 胰腺癌组织标本中有较高的端粒酶活性表达；胰液脱落细胞检测端粒酶活性无助于鉴别胰腺良恶性疾病。

Telomerase activity in pancreatic carcinoma specimen and pancreatic juice cell

Aim Most cancers and cell lines have detectable telomerase activity,so it is regarded as a promising tumor marker.In order to detect pancreatic neoplasm in early stage,we tried to detect telomerase activity in pancreatic carcinoma specimens and pancreatic juice cells collected during ERCP.Methods 6 pancreatic cancer cell lines,14 surgical specimens and 27 pieces of pancreatic juice were detected.Pancreatic juice was collected during ERCP after injection of 100 cu secretin.TRAP ELISA was used to detect telomerase activity.Results All 6 pancreatic cancer cell lines had detectable telomerase activity.63%(5/8) of the pancreatic carcinoma specimens had detectable telomerase activity,while 2 benign pancreatic disease and 4 normal pancreatic tissues showed no telomerase activity.As for centrifuged pancreatic juice cells,58%(11/19) of the pancreatic carcinoma and 26%(5/8) of the benign pancreatic diseases had detectable telomerase activity.Conclusions Although telomerase activity is higher in the specimens of pancreatic carcinoma than that those of benign and normal cases,telomerase activity has no significance in the pancreatic juice cells of pancreatic carcinoma and benign pancreatic diseases.Detection of telomerase activity in pancreatic juice cells is not sensitive and specific to diagnose pancreatic carcinoma.
