| 摘 要： | 探讨５－氟脲嘧啶诱导人胃癌细胞凋亡的规律及在胃癌化疗中的意义。方法：应用细胞形态观察,琼脂糖凝胶电脉,流式细胞仪检测和观察５－ＦＵ对胃癌细胞ＳＧＣ－７９０１增殖周期的影响以及杀伤细胞的方式,并检测了ＳＧＣ－７９０１细胞表面ｂｃｌ－２、ｂａｘ蛋白的表达情况。

| Abstract: | Objective: To observe the pattern of 5-fluorouracil-induced apoptosis of the cells of gastric cardinoma cell line and eva1uate the role of the drug in the chemotherapy of the malignancy. Methods: The effects of 5-fluorouracil on the growth cycle and apoptosis of the cells of SGC-7901 cell line were observed with cell morphology, electrophoresis on agarose gel and flow cytometry and the expression of bcl-2 and bax protein in the carcinomatous cells also examined. Results: 5-fluorouracil induced significant apoptosis of SGC-7901 cells and internucleosomal DNA fragmentation displayed the chracteristic ladder-lide .arrangement of the electrophoretic bands. The expression of bcl-2 was downregulated. Conclusion: It is possible that 5-fluorouracil exercises its chemotherapeutic effects on gastric carcinoma through its induction of cell apoptosis. |
