补肾健脾方对AD大鼠AchE、ChAT及海马神经元凋亡的影响

摘    要:目的研究中药补肾健脾方对阿尔茨海默(AD)大鼠血清及海马组织中乙酰胆碱酯酶(AchE)、胆碱转移酶(ChAT)及神经细胞凋亡的影响。方法雄性SD大鼠随机分为5组:正常组、模型组、假手术组及补肾健脾复方制剂小剂量、大剂量干预组。采用鹅膏蕈氨酸毁损大鼠脑Meynert基底核制备AD动物模型,中药补肾健脾方分别以3.6g/(kg.d)、7.2g/(kg.d)灌胃。30d后以水迷宫测试大鼠学习及记忆能力,对各组大鼠血清及脑海马匀浆中AchE、ChAT的活性进行测定,运用电镜技术及免疫组化检测海马中神经细胞的凋亡及Bax、Bcl-2的表达。结果与模型组比较,补肾健脾复方制剂大、小剂量干预组大鼠的平均逃避潜伏期明显缩短(P〈0.05),血清及匀浆中AchE的活性下降(P〈0.05),ChAT活性增高(P〈0.05),脑皮质的细胞凋亡显著减少(P〈0.05),脑皮质中Bax阳性神经元数量明显降低(P〈0.05),Bcl-2阳性神经元的数量显著增加(P〈0.05)。结论补肾健脾方可在抑制AchE活性的同时增加ChAT的活性,并可通过调节Bax/Bcl-2的表达减少海马组织中神经细胞的凋亡。

Effect of Bushen Jianpi Recipe on AchE and ChAT Activity and Hippocampal Neurons Apoptosis in Rats

Abstract:Objective To investigate the effect of Bushen Jianpi recipe(BJR) on activities of acetylcholinesterase(AChE)and choline acetyltransferase(ChAT)and hippocampal apoptosis in rats with Alzheimer’s disease.Methods Imitate Alzheimer’s diseast models in rats were established by ibotenic acid.Rats were randomly divided into five groups:Control group,model group,sham operation group,BJR low dose group treated by gavage with BJR at the dosage of 3.6 g/(kg·d),and high dose group treated by gavage with BJR at the dosage of 7.2 g/(kg·d).The learning and memory function of rats were evaluated with water maze test.The activity of AchE,ChAT and hippocampal apoptosis were determined.The expression of Bax and Bcl-2 were detected by immunohistochemical staining.Results Compared with model group,the escape latency were significantly shortened in BJR low and high dose group(P0.05).The activity of AchE was decreased in blood serum and hippocampal homogenate,while the activity of ChAT was increased in BJR group(P0.05).The hippocampal neurons apoptosis was decreased(P0.05),the expression of Bax protein was decreased(P0.05),and the expression of Bcl-2 protein was increased in BJR group(P0.05).Conclusion Bushen Jianpi recipe could inhibit the AchE activity,augment the ChAT activity,and inhibits hippocampal neurons apoptosis by regulating the expression of Bax/Bcl-2 protein in rats with Alzheimer’s disease.
