FAK在食管癌组织中的表达及临床意义

摘    要:
目的检测局部黏着斑激酶(focal adhesion kinase,FAK)在食管癌组织及正常食管组织中的表达,探讨FAK在食管癌发生、发展中的意义。方法采用免疫组化SP法检测FAK在40例食管癌组织、40例正常食管组织中的表达情况。结果免疫组化结果提示:40例食管癌组织中有16例(40.0%)FAK呈强阳性染色,15例(37.5%)FAK呈弱阳性染色,9例(22.5%)FAK表达缺失;40例正常食管组织中,3例(7.5%)FAK呈强阳性染色,7例(17.5%)FAK呈弱阳性染色,30例(75.0%)FAK表达缺失,FAK在食管癌组织的阳性表达率(77.5%)显著高于正常食管组织的阳性表达率(25.0%),两组比较差异有统计学意义(P0.05)。FAK的表达与食管癌患者的性别、年龄无关(P0.05),而与分化程度相关(P0.05),低分化食管癌组FAK蛋白表达高于高中分化组。结论 FAK在食管癌组织中表达增高,其表达量与食管癌的分化程度有关,FAK在正常食管组织中表达缺失或减少。FAK可能与食管癌的发生、发展有重要关系,其表达的高低有可能作为食管癌预后判断的一种新指标。

Abstract:
Objective To study the expression of FAK in the tissues of esophageal carcinoma and normal esophagus, and investigate its clinical significance. Methods The expression of FAK protein was detected by immunohistochemis- try SP method in the tissues of esophageal carcinoma (40 cases) and normal esophagus (40 cases). Results Immuno- histochemistry: among 40 cases of esophageal carcinoma, FAK was strongly positive staining in 16 cases (40.0%) , 15 cases (37.5 % ) showed a weak positive staining of FAK, there was no expression of FAK in 9 cases (22.5 % ). Among 40 cases of normal esophagus, FAK was strongly positive staining in 3 cases (7.5%), 7 cases ( 17.5% ) showed a weak positive staining of FAK, there was no expression of FAK in 30 cases (75.0%) , the expression rate of FAK in e- sophageal carcinoma was significantly higher than that of normal esophagus and the difference had statistically signifi- cance (P 〈 0.05 ). The expression of FAK was related with cellular differentiation degree ( P 〈 0.05 ) , not related with the gender and age (P 〉0.05). Conclusion The expression of FAK is higher in the tissue of esophageal carcinoma, which is related with cellular differentiation degree. In the tissue of normal esophagus, the expression of FAK protein is absent or decreased. FAK is associated with the development and progression of esophageal carcinoma. FAK may be taken as a new marker in evaluating the prognosis of patients with esophageal carcinoma.
