摘    要:目的 探讨聚合胶原对肌成纤维细胞表型转化的促进作用及其可能机制.方法 用聚合胶原培养小鼠成纤维细胞,观察聚合胶原对成纤维细胞形态学及生长曲线影响.蛋白印迹法分析PTEN及Akt Ser473及p42/44磷酸化水平的情况.结果 在聚合胶原中培养的成纤维细胞可以向肌成纤维细胞转化.在聚合胶原的诱导下,成纤维细胞内PTEN含量减少,磷酸化Akt增多,同时细胞内磷酸化p42/44MAPK含量增多.结论 聚合胶原可以诱导成纤维细胞向肌成纤维细胞转化.其机制可能与PTEN和p42/44MAPK信号通路相关.

Abstract:Objective To study the effect and potential mechanism of polymerized collagen on myofibroblast differentiation.Methods The cardiac fibroblasts were cultured with polymerized collagen in vitro in mice.Morphological changes and growth curve of cardiac fibroblasts were observed.Expressions of PTEN,p-Akt Ser473 and p-p44/42 MAPK(ERK1/2) were measured by Western blot.Results Normal cardiac fibroblasts cultured with polymerized collagen were able to differentiate into myofibroblasts.Induced by polymerized collage...
