人参黄芪含药血清对人脐静脉内皮细胞活性的影响

摘    要:目的:通过观察养心汤中君药(人参、黄芪)对人脐静脉内皮细胞增殖率的影响,探讨其防治冠心病不稳定型心绞痛的作用机制。方法:实验细胞分为空白血清组、模型组、参芪低剂量组、参芪中剂量组、参芪高剂量组5组。应用MTT法检测细胞活性。结果:H2O2培养液可使人脐静脉内皮细胞收缩变圆,细胞间隙增宽,部分细胞脱落。随参芪含药血清浓度的增加,细胞越成片贴壁生长,细胞形态和间隙越接近正常。经MTT法检测,与空白血清组相比,模型组增值率明显降低,差异具有统计学意义(P0.05);各参芪组间比较,参芪高剂量组增殖率显著升高,差异具有统计学意义(P0.05)。结论:参芪含药血清能预防H2O2对人脐静脉内皮细胞造成的损伤,并能够促进内皮细胞增殖,这可能是其防治冠心病不稳定型心绞痛的作用机制之一。

Abstract:Objective : To observe the proliferation rate which affects human umbilical vein endothelial cell. disease. Methods: The HUVEC were divided low - dose Shenqi group, middle - dose Shenqi using MTT assay. Results: The cell shrinkage by the drug ( Ginseng, AstragahJs) of Yangxin decoction This study is to investigate the possible mechanism of UA into five groups. They were blank serum group, model group, group and high -dose Shenqi group. Detect the cell activity by and cell gap were obviously enlarged in model group. With the increased drug concentrations, the cells were more liable to adherent grow in fiat polygonal shape, intercellular closer to normal. The MTT results showed that compared with the blank serum group, the OD value was significantly lower in model group (P 〈 0. 0:5 ) ; Between two Shenqi groups, Shenqi group with high dose was higher than others(P 〈 0. 05 ). Conclusion: The Ginseng and Astragalus combination serum have a clear preventive effect on the model using H202 injury HUVEC, play an important role in the treatment of UA. which can promote proliferation of endothelial cells.
