法舒地尔对家兔脊髓缺血再灌注损伤的影响

摘    要:
目的观察法舒地尔对家兔脊髓缺血再灌注损伤后细胞间粘附分子-1(ICAM-1)表达水平的影响。方法家兔42只,随机分为假手术组、模型组、法舒地尔组,采用腹主动脉夹闭法制作缺血再灌注模型,法舒地尔组每日腹腔注射法舒地尔,模型组注射等量生理盐水,假手术组不予干预。手术后采用Jacobs法对家兔后肢功能评级,肌电图检测后肢运动诱发电位。免疫组化法观察脊髓缺血区ICAM-1的表达水平。结果模型组ICAM-1表达明显,后肢瘫痪严重,法舒地尔能显著抑制ICAM-1的表达,瘫痪程度较轻,后肢运动诱发电位潜伏期缩短。结论法舒地尔可能通过抑制Rho激酶活性,进而抑制ICAM-1的表达,减轻脊髓缺血再灌注损伤的炎性反应,达到保护作用。

Effect of Fasudil on Spinal Ischemia-reperfusion Injury in Rabbits

Abstract:
Objective To observe the effect of Fasudil on rabbit spinal ischemia-reperfusion(I/R) injury of the expression of intercellular adhesion molecules-1(ICAM-1),and to explore the mechanism. Methods 42 Rabbits were divided into sham group,control group,Fasudil treatment group randomly.The I/R model of the spinal cord was conducted by the selective occlusion of the abdominal aorta.The treatment group was injected with Fasudil through abdominal cavity every day,while the control group was injected equivalent physiological saline in the same way.The sham group was not injected everything.The function of hindlimbs was evaluated by the Jacobs method.In addition,the motor evoked potentials(MEP) of hindlimbs was dectected through myoelectricity graph.The expression of ICAM-1 in spinal ischemia was detected through immunohistochemistry. Results The expression of ICAM-1 in the control group was increased remarkably,and the paralysis of hindlimbs was seriously.The expression of ICAM-1 was restrained remarkably after treated with Fasudil,and the paralysis of hindlimbs was lighter.The incubation period of MEP in the hindlimbs was more shortened.The statistic significance was remarkable. Conclusions Fasudil provides a neuroprotective effect on rabbits following spinal I/R injury through relieving inflammatory reaction,and restraining the expression of ICAM-1.
