胃癌p53基因甲基化及其表达情况的研究

摘    要:
目的检测胃癌组织中p53基因甲基化状态和p53蛋白表达情况。方法选择p53基因第5外显子,应用限制性内切酶HpaII和MspI酶切胃癌组织及正常胃组织DNA,经PCR扩增和免疫组化等方法对100例胃癌患者的癌组织和癌旁组织进行检测。结果有71%的患者p53蛋白表达阳性,54%的患者具有p53蛋白基因的高甲基化,50%同时有p53蛋白表达阳性和p53基因的高甲基化。结论提示p53基因高甲基化状态引起的突变或失活是胃癌发生发展的重要因素之一。

Abstract:
Objective To research the abnormalities of p53 gene expression and the methylation status of p53 in primary gastric cancer. Methods The expression of p53 protein was detected with immunohistochemistry. The methylation of p53 gene were measured by using PCR-based restriction endonuclease enzymes HpaII and MspI in 100 samples of gastric cancer and 100 samples of corresponding normal gastric tissues. Results 71% of gastric cancer samples have positive staining in p53 protein, 54% of gastric cancer samples have hyper- methylation of p53 protein, 50% of the gastric cancer samples have positive staining and hypermethylation at the same time. Conclusion The data suggested that the gene mutation and inactivation induced by p53 gene hypermethylation status play important roles in the genesis and progress of cancer of stomach.
