斜链拟青霉胞外多糖对免疫抑制小鼠免疫功能的影响

摘    要:目的:探讨斜链拟青霉胞外多糖对免疫抑制小鼠免疫功能的调节作用。方法:随机选取6只小鼠作为正常对照组,其余24只小鼠连续3 d腹腔注射环磷酰胺80 mg/kg,造模后随机分为4组,其中3组分别进行腹腔注射斜链拟青霉胞外多糖100 mg/kg、300 mg/kg、900 mg/kg,而模型对照组及正常对照组给予等量生理盐水,各组连续干预7 d,末次给药12 h后处死,计算脾湿质量指数、腹腔巨噬细胞吞噬指数;检测小鼠脾淋巴细胞增殖活性和IL-12含量。结果:模型组小鼠上述各项免疫指标均有所下降,相对模型组,多糖给药组小鼠脾湿质量、吞噬指数、脾淋巴细胞的增殖活性及其分泌的IL-12显著提高(P<0.05)。结论:斜链拟青霉胞外多糖对外源性免疫抑制剂所致免疫功能抑制具有增强效果。

Abstract:Objective:To investigate the effects of Paecilomyces cateniobliquus polysaccharide on immune function induction in immunosuppressed mice.Methods:Six mice were randomly selected as normal control group,and twenty-four other mice were induced to establish immunosuppressive model by the peritoneal injection of cyclophosphamide(80 mg/kg) for three consecutive days.After model preparation,the immunodeficiency mice were divided into four groups in which three were injected with Paecilomyces cateniobliquus polysaccharide solution in dose of 100 mg/kg,300 mg/kg and 900 mg/kg,respectively.Animals in model and normal control groups were daily injected with equal physiological saline for 7 successive days and were killed in 12 hours of last administration.Wet weight index of spleen and phagocytic index were calculated.Activities of thymus cell and the level of IL-12 in mice were measured.Results:The above indexes for the mice in model group were decreased,and the wet weight index of spleen,phagocytic index,the thymus cell proliferation and IL-12 production were increased significantly in polysaccharide groups as compared with the model group(P<0.05).Conclusion:Paecilomyces cateniobliquus polysaccharide has a better effect on enhancing immunocopetence in immunosuppressive model mice caused by cyclophosphamide.
