辐照含银同种异体神经移植的实验研究

摘    要:
目的观察辐照含银处理同种异体神经的体外抑菌效果及移植效果。方法①将同种异体神经分别行辐照含银、单纯辐照和低温冷冻处理,并对它们进行含银量测定、细菌培养及体外抑菌实验。②18只健康成年家兔,随机分为3组,分别移植辐照含银同种异体神经(A组)、新鲜异体神经(B组)和自体神经(C组)。通过观察移植后宿主的免疫排斥反应、组织形态学和光镜下组织结构变化来判断移植效果。结果①辐照含银处理神经含Ag^+(193.2±10.3)μg/g;辐照含银神经组具有明显抑菌作用,与其他各处理组相比差别显著(P〈0.01);②细胞毒实验显示A组淋巴细胞死亡率与C组无明显差异(P〉0.05);术后12周,A组与C组腓肠肌萎缩好转,针刺可见回缩反应;光镜下可见雪旺细胞增生,新生轴索出现,轴索数量增加,B组移植神经溶解。结论辐照含银同种异体神经能解决降低抗原性、组织保存、神经再生效果和机能恢复、术后感染等问题,是较理想的神经移植材料。

Abstract:
Objective To observe the in vitro bacteriostatic and transplantation effect of irradiated allogenic nerve containing silver. Methods ① Allogenic nerve was treated with irradiation after bathed with compound silver,simple irradiationand hypothermal freeze respectively. Also each nerve treated with the method above was processed with silver content determination, bacterial culture and vitro bacteriostatic experiment. ②18 healthy adult rabbits were randomly divided into three groups. A, B and C. Each group ning silver, fresh allogenic nerve, and plated immunological rejection of the was transplanted with irradiated homologous nerve contaiautogeneic nerve respectively. We observed the plst-transhost and judged the transplantarion effect by histomorphology and the changes of tissue structure under light microscope. Results The content of silver in group A was (193.2±10.3)μg/g. The in vitro bacteriostatic experiment of group A exhibited a remarkable bacteriostatic effect and showed significant difference compared with the other two groups(P〈0. 01). At 12 weeks after the treated nerve was transplanted in group A and C,we could see bettered atrophy of the gastrocnemius,and recovery response after acupunctured. Also, we could see the proliferation of the Schwann cell and the occurrence of the regenerated anxiliary fibers. The transplanfed nerve in Group B dissolved. Conclusion Irradiated allogenic nerve containing silver could solve the problems such as. reduction of the antigenicity, tissue conservation, nerve regeneration,function recovery,postoperative infection, et al. Also, it is a comparatively ideal material for nerve transplantation.
