藤梨根提取物对人胃癌BGC细胞凋亡及Bax蛋白表达的影响

摘    要:目的:探讨藤梨根提取物对人胃癌BGC-823细胞凋亡的影响及其调控机制。方法:体外培养对数生长期胃癌BGC-823细胞,采用不同浓度的藤梨根提取物作用于人胃癌BGC-823细胞,流式细胞仪检测细胞凋亡,采用免疫组织化学方法检测Bax蛋白表达。结果:藤梨根作用于BGC-823,在8~32mg·mL^-1时呈不同程度的促进凋亡;藤梨根提取物大、中、小剂量组对人胃癌BGC-823细胞中Bax表达水平上调。结论:藤梨根提取物对人胃癌BGC-823细胞的增殖具有明显抑制作用,在实验浓度范围内呈浓度依赖性促进细胞凋亡,这可能与激活Bax蛋白表达有关。

Abstract:Objective:To discuss the effects of Kiwi root extract on human gastric cancer BGC cell apoptosis and its regulatory mechanism. Methods:The gastric cancer BGC-823 cells in logarithmic phase were cultivated in vitro. Kiwi root extract with different concentrations act on gastric cancer BGC-823 cells. The cell apoptosis was detected by Flow cytometry instrument, and Bax protein expression was tested by immunohistochemical methods. Results: Kiwi root can act on BGC-823 cells. Kiwi root in 8- 32mgoml^- 1 can promote cell apoptosis in the different level. Kiwi root extract raised Bax expression levels of human gastric cancer BGC- 823 cells in large dose group, medium dose group or small dose group. Conclusion.. Kiwi root extract has obvious inhibitory effecton human gastric cancer BGC-823 cells proliferation. Kiwi root extract in the experimental concentration range can promote cell ap optosis in concentration dependence, which may be related to activation of Bax protein expression.
