肝门阻断对大鼠肠肌间神经丛内AchE阳性神经元的影响

摘    要:
目的:研究肝门阻断后大鼠肠肌间神经丛内乙酰胆碱酯酶(AchE)阳性神经元表达的变化规律,以探讨肝门阻断对肠道功能影响的神经机制.方法:SD大鼠分成实验组(再按阻断时间分为肝门阻断20 min组、40 min组和60 min组)及对照组,取相同部位的部分小肠和结肠,制作肠肌间神经丛铺片标本行AchE染色,观察和比较各组AchE阳性神经元的分布密度和染色情况.结果:各实验组与对照组比较,肠肌间神经丛内AchE阳性神经元数量减少(P<0.05或P<0.01),且AchE阳性表达减弱;各实验组比较,肝门阻断60 min组的AchE阳性神经元数量明显少于20 min组(P<0.05),20 min组的AchE阳性神经元胞体内可见强阳性染色颗粒,40 min组和60 min组不易见强阳性染色颗粒,染色较浅.结论:肝门阻断会影响或损伤肠神经系统的胆碱能神经,这可能是术后肠道运动功能障碍发生的神经机制.

Abstract:
Objective: To study the changes of AchE positive neurons expression in myenteric plexus of rat intestinal tract after portal triad clamping and to explore the nervous mechanism of the effects of portal triad clamping on intestinal tract function.Methods: SD rats were divided into experimental groups(portal triad clamping 20 min group,40 min group,and 60 min group) and control group,and specimens of intestinal myenteric plexus were made from small intestines and colons and stained with AchE histochemistry method.Results: Compared with control group,the numbers of AchE positive neurons in intestinal myenteric plexus decreased in experimental groups(P<0.05 orP<0.01),and the AchE positive expression decreased;in experimental groups,AchE positive neurons are fewer in 60 minutes group than in those 20 minutes group(P<0.05)and strong positive granules could be seen in AchE positive neurons in 20 minutes group,but it was difficuit to be found in 40 and 60 minutes group.Conclusion: Intestinal motor dysfunction resulted from portal trial clamping is related to injuries of cholinergic nerve in enteric nervous system.
