凉血化瘀方对MRL/lpr狼疮鼠蛋白尿及抗ds-DNA抗体的影响

[目的]研究凉血化瘀方对MRL/lpr狼疮鼠蛋白尿及抗ds-DNA抗体的影响。[方法]MRL/lpr小鼠35只,饲养至3月龄。将35只雌性MRI/lpr小鼠随机分为五组,每组7只。分别为:阴性组、模型组、西药强的松加环磷酰胺组、中药加强的松加环磷酰胺组、中药组。实验小鼠均于灌胃前、灌胃21d、灌胃49d时代谢笼采集24h尿蛋白定量;眼眶后静脉丛采血,取血并分离血清后,置-70℃冰箱保存,行ELISA检测抗ds-DNA抗体的表达。[结果]MRL/lpr小鼠模型组ds-DNA水平较阴性组明显升高,蛋白尿高于阴性组。经过西药(CTX、强的松)、中药(凉血化瘀方)及中西医结合治疗(CTX、强的松、凉血化瘀方),各治疗组蛋白尿显著少于模型组。但降蛋白尿上,西药组疗效优于中药组,中西医结合组疗效优于单用中药或单用西药组,差异有统计学意义(P<0.05)。抗ds-DNA抗体血清含量模型组3.026±0.129,阴性组1.404±0.064,两组相比,P<0.01。[结论]中药凉血化瘀方具有降低血抗ds-DNA水平及蛋白尿的作用。

Effect of Formula with Function of Cooling Blood to Remove Blood Stasis on Albuminuria and Anti - Dsdna Antibodies in MRL/Lpr SLE Mice

[ Objective ] To study the effect of formula with function of cooling blood to remove blood stasis on albuminuria and anti - dsDNA antibodies in MRL/lpr SLE mice. [ Methods ] From 3 months of age, 35 female MRL/Ipr mice were randomly divided into control group, model group, Western medicine （combination of prednisone and cyclophosphamide） group , combination of Chinese and Western medicine group, Chinese medicine group. 7 mice were included in each group . The 24 - hour urine protein of experimental mice was respectively tested before intragastric administration, 21 and days 49 after intragastric administration. Blood was collected from the venous plexus. Serum were segregated and stored in -70~C refrigerator. The expres- sion of anti -dsDNA antibodies were determined with enzyme linked immunosorbent assay（ELISA）. [ Results ] The ds - DNA levels of MRL / lpr mice model group were significantly higher than those of the control group, the proteinuria level was slso higher than that of the contrasting group. After therapy, proteinuria in all treated groups was significantly less than that in model group. However, the proteinuria reduction of the Western med- icine group was superior to the Chinese medicine group （ P 〈 0.05 ） , and that of the combination of Chinese and Western medicine group was superior to the Western medicine group and Chinese medicine group （P 〈 0. 05 ）. The ds- DNA level was 3. 026 ± 0. 129 in the model group and 1. 404 ± 0. 064 in the control group. The difference was Statistically significant（ P 〈0.01 ）. [ Conclusion] Formula with function of cooling blood to remove blood stasis can lower the level of albuminuria and anti - dsDNA antibodies.
