摘    要:目的:研究As2O3对肝癌细胞株HLE细胞毒和诱导凋亡作用。方法:应用MTT染色,流式细胞仪和电子显微镜观察As2O3对HLE细胞株的诱导凋亡作用和形态学改变,应用共聚焦激光扫描显微镜(CLSM)测试肝癌细胞内[Ca^2 ]i变化。结果:As2O3对肝癌细胞株HLE具有诱导凋亡作用,CLSM检测肝癌细胞内[Ca^2 ]i高浓度As2O3作用下48h达最高峰;在低浓度下96h达高峰。结论:As2O3对肝癌细胞具有较强的细胞毒和诱导细胞凋亡的作用。

Abstract:Objective To study the effects of cytotoxicity and induction of apoptosis by arsenic trioxide(As 2O 3) on hepatoma cell line HLE.Methods The effect of cytotoxicity by As 2O 3 on hepatoma cell lines HLE was determined by MTT assay, apoptosis and morphologic changes of HLE induced by As 2O 3 by electronic microscope and flow cytometry. The intracellular Ca 2+ concentration was detected by confcal Laser scanning microscope(CLSM).Results As 2O 3 could induced apoptosis in hepatoma cell lines remarkably, HLE cells exhibited typical morphologic features of apoptosis. The intracellular Ca 2+ concentration reached peak 48 hours after treatment with high concentration of As 2O 3, and 96 hours after treatment with low concentration of As 2O 3. Conclusions As 2O 3 exhibited significantly cytotoxicity and induction of apoptosis on hepatoma cell lines HLE.As 2O 3 might be promising in the treatment of hepatic carcinoma, and be worthy to be further studied.
