PTEN基因细胞间黏附分子与肝细胞发生发展侵袭转移关系的研究

摘    要:目的探讨PTEN基因和ICAM-1与肝细胞发生、发展、侵袭转移的关系及临床意义。方法应用组织芯片技术和免疫组化SABC法对46例手术切除的经病理确诊为肝细胞癌变的标本进行实验研究。结果正常肝细胞组织中PTEN有4例呈强阳性表达,ICAM-1均呈阴性表达。肝细胞癌变患者PTEN基因阳性率为65.2%(30/46),而ICAM-1阳性率为58.7%(27/46),癌旁组织中PTEN基因和ICAM-1阳性率分别为90.9%(30/33),87.8%(29/33),两者表达与患者年龄、性别、肿瘤大小及AFP是否阳性无关,而与肝细胞癌变的病理分级,包膜及癌栓的有无关系密切。其中PTEN与HBsAg是否阳性无关,ICAM-1与HBsAg是否阳性关系密切。结论 PTEN基因具有抑制肝细胞的癌变发生、发展、侵袭转移的作用,而ICAM-1对肝细胞的癌变、发生、发展、侵袭转移具有促进作用。本研究发现两者在肝细胞的癌变、发生、发展、侵袭转移过程中两者呈负相关。

Abstract:OBJECTIVE To explore the PTEN gene and ICAM and hepatocellular carcinoma occurrence,development,invasion and metastasis and its clinical significance.METHODS The tissue microarray technique and immunohistochemical SABC method was used for the pathological section of 46 cases.RESULTS Patients with HCC PTEN gene and ICAM-1 positive rates were 65.2%(30/46)and 58.7%(27/46),respectively.The carcinoma tissue and the positive rates were 90.9%(30/33)and 87.8%(29/33),respectively.Both expression had no association with age,gender,tumor size and AFP,and had no closely association with hepatic cell carcinoma pathological grading,envelope and cancer.The PTEN had no association with HBsAg,while the ICAM-1 had significantly association with the HBsAg.CONCLUSION The PTEN gene is negative correlated with ICAM-1 in HCC occurrence,development,metastasis and invasion.
