姜黄素对高脂饮食大鼠血脂含量和caveolin-1表达的影响

摘    要:目的观察姜黄素对高脂饮食大鼠总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量和caveolin-1表达的影响。方法将40只健康成年雄性大鼠分为正常对照组(普通饲料喂养)、高脂血症模型组(高脂饲料喂养)、姜黄素低剂量组(高脂饲料+姜黄素100mg/kg.bw/d)、姜黄素高剂量组(高脂饲料+姜黄素200mg/kg.bw/d)。分别于实验开始前1d、实验第8周末禁食12h后,测血清TC、TG、LDL-C含量的变化。同时Western-Blot印迹法检测大鼠血管壁caveolin-1表达的变化。结果与正常对照组比较,高脂血症模型组的TC、TG、LDL-C分别由对照组的(1.58±0.25)、(0.55±0.06)、(0.49±0.06)mmol/L升高到高脂饮食组的(3.47±0.35)、(0.92±0.07)、(0.88±0.04)mmol/L,并伴随caveolin-1蛋白表达水平下降3.25倍。姜黄素的介入能降低高脂饮食大鼠TC、LDL-C、TG的含量以及促进其caveolin-1的表达(P0.05)。结论姜黄素具有降低高脂饮食大鼠TC、LDL-C、TG含量的作用,其作用机制可能与促进caveolin-1表达有关。

Effect of Curcumin on Serum Lipids and Caveolin-1 Expression in High-fat-fed Rats

Abstract:Objective To observe the effect of curcumin on the content of TC,TG,LDL-C,and caveolin-1 expression in high-fat-fed rats.Methods Forty adult male rats were randomly divided into four groups:control group,hyperlipidemic group (high-fat-fed),low-dose curcumin group (high-fat-fed plus curcumin,100 mg/kg.bw/d),and high-dose curcumin group (high-fat-fed plus curcumin,200 mg/kg.bw/d).The levels of serum total cholesterol (TC),total triglyceride (TG),and low density lipoprotein-cholesterol (LDL-C) were measured after 8 weeks of experimental treatment.Western-Blot analysis was used to detect caveolin-1 protein expression in aortic wall of experimental rats.Results As compared with control group,serum lipids significantly increased in high-fat-diet group,such as TC changing from (1.58±0.25) mmol/L to (3.47±0.35) mmol/L,TG changing from (0.55±0.06) mmol/L to (0.92±0.07) mmol/L,LDL-C changing from (0.49±0.06) mmol/L to (0.88±0.04) mmol/L.At the same time,the level of caveolin-1 protein expression decreased by 3.25 times in high-fat-diet group.These serum lipid levels were decreased in curcumin treated groups (P〈0.05).Conclusions Curcumin could decrease the levels of serum lipids in high-fat-fed rats,and the molecular mechanism might be associated with up-regulation of caveolin-1 expression.
