人胃癌裸鼠原位移植和转移模型的建立及三种方法比较

摘    要:目的:建立胃癌裸鼠原位移植和转移模型,并比较三种不同方法的结果。方法:将胃癌肿瘤细胞悬液、胃癌裸鼠皮下移植瘤块和胃癌裸鼠原位移植瘤块种植于裸鼠胃壁,形成原位移植瘤,观察和比较三种方法所建立的模型肿瘤生长状况、移植成功率和自发转移的发生率。常规HE染色,观察胃癌原位移植瘤、胃癌淋巴结转移和肝转移的病理切片。结果:细胞悬液种植的原位胃癌成瘤率为50%,淋巴结广泛转移率为37.5%,肝转移发生率为25%;皮下瘤块的原位成瘤率为100%,淋巴结广泛转移率为62.5%,肝转移发生率为50%,腹水形成率为6.25%;而胃癌裸鼠原位移植块再次原位移植的原位成瘤率达100%,淋巴结广泛转移率为87.5%,肝转移发生率为75%,腹水形成率为12.5%。细胞悬液成瘤时间较皮下肿瘤组织块和胃癌裸鼠原位移植块再次移植延迟2—3周,而后两者无差异。结论:三种原位移植和转移模型均具有人胃癌自然生长过程的特点,以胃癌裸鼠原位移植块再次移植为最优,且更具有人胃癌的生物学活性。

Abstract:Objective: To develop three methods of orthotopic im plant and metastatic models of human gastric cancer in nude mice and to compare these methods. Methods: Tumor cell suspension, intact subcutaneous ly tumor tissue or orthotopically gastric tumor tissue were orthotopically impla nted into the nude mouse stomach to develop implanted tumors, The tumor growth c haracteristic, tumor taking rates and metastatic rates of the three models were analyzed and compared. Observe the gastric orthotopically transplantation, gastr ic cancer lymphatic metastasis and hepatic metastasis by HE staining.Resu lts: In the model implanted with tumor cell suspension, the local tumor g rowth was slower,tumor taking rate was 50%, The metastases of lymph nodes and l ivers were only 37.5% and 25%. In the model implanted with intact subcutaneou sly tumor tissue, tumor taking rate was 100%, the metastases of lymph nodes and livers were 62.5% and 50%, ascite formation is 6.25%. In the model implant ed with orthotopically gastric tumor tissure, tumor taking rate was 100%, the m etastases of lymph nodes and livers were 87.5% and 75%, ascite formation is 1 2.5%. Conclusion: There were growth characteristics of human sto mach cancer in all orthotopic tumor models. The model implanted with orthotopica lly gastric tumor tissue owned the biologic activity best than the others. [
