| Abstract: | Objective:To studywhethermousesertolicellscaninducetheapoptosisof maturedT lymphocytesincocultures in vitro.Methods:WithTUNEL,DNAelectrophoresis,electro-micrographyandflowcytometry,we examinedtheapopto-sisanditsratesof mousematuredT lymphocytesincontrolgroup(T lymphocytesonly),groupA(T lymphocytes+culture mediumof sertolicells),groupB(T lymphocytes+sertolicells).Results:Underelectro-micrography,chromatinconden-sation,karyopyknosis,karyorhexisandapoptoticbodywereobservedinsomeT lymphocytesin3groups;somenucleuses werestaineddarkbluewithTUNEL;a typicalDNAladderwas foundwithDNAelectrophoresis.Theapoptoticratesof T lymphocytesingroupA andB weresignificantlyhigherthanthatincontrolgroup(P<0.01).Theapoptoticrateof T lym-phocytesingroupB wassignificantlyhigherthanthatingroupA(P<0.01).Conclusion:Incocultureconditionin vitro,mousesertolicellscaninducetheapoptosisof maturedT lymphocytes. |
