MTSS1在胃癌组织中的表达及对胃癌细胞增殖凋亡的作用研究

摘    要:
目的 探讨肿瘤转移抑制候选基因(MTSS1)在胃癌组织中的表达水平及对胃癌细胞增殖凋亡的作用.方法 Western Blot检测胃癌组织及对应的癌旁组织中MTSS1蛋白表达水平.通过细胞转染的方法 将过表达载体pEGFP-N1-MTSS1、空载体pEGFP-N1转染至胃癌细胞中,MTT法检测转染48 h的细胞增殖情况,流式细胞术检测转染48 h的细胞凋亡情况,Western Blot检测转染48 h细胞中Cleaved Caspase-3、HES1、NICD1、NICD2蛋白表达水平.胃癌细胞与NOTCH信号通路抑制剂S2188作用48 h后,检测细胞凋亡及细胞中Cleaved Caspase-3、HES1、NICD1、NICD2蛋白表达水平.结果 胃癌组织中MTSS1蛋白水平明显低于癌旁组织(P=0.000);pEGFP-N1-MTSS1组细胞存活率明显低于pEGFP-N1组(P=0.000);pEGFP-N1-MTSS1组细胞凋亡率明显高于pEGFP-N1组(P=0.000);pEGFP-N1-MTSS1组细胞中Cleaved Caspase-3、HES1、NICD1、NICD2表达水平与pEGFP-N1组比较,差异均有统计学意义(P=0.000);NOTCH信号通路抑制剂S2188作用胃癌细胞48 h后,抑制剂组的细胞凋亡率明显高于对照组(P=0.000),两组的Cleaved Caspase-3、HES1、NICD1、NICD2蛋白表达水平比较,差异均有统计学意义(P=0.000).结论 MTSS1在胃癌组织中表达下调.MTSS1能促进胃癌细胞凋亡,抑制胃癌细胞增殖,作用机制与NOTCH信号通路有关.

Abstract:
Objective To investigate the expression of MTSS1 in gastric cancer and its effect on the proliferation and apoptosis of gastric cancer cells. Method The expression level of MTSS1 was detected in cancer tissues and adjacent tis-sues by Western Blot. The expression vector pEGFP-N1-MTSS1 and plasmid pEGFP-N1 were transfected into gastric cancer cells, in which the proliferation was detected by of MTT after 48 h, and apoptosis by flow cytometry, the expres-sion levels of Cleaved Caspase-3, HES1, NICD1, NICD2 protein in cells were detected by Western Blot. Gastric cancer cells were treated with NOTCH signal pathway inhibitor S2188 for 48 h, and the cell apoptosis and the expression levels of Cleaved Caspase-3, HES1, NICD1, NICD2 protein in cells were analyzed. Result The level of MTSS1 protein in gas-tric cancer tissues was significantly lower than that in adjacent tissues (P=0.000);the survival rate was significantly lower (P=0.000), while the apoptosis rate was significantly higher in pEGFP-N1-MTSS1 group compared with pEGFP-N1 group (P=0.000); besides, the expression of pEGFP-N1, Cleaved Caspase-3 HES1, NICD1, NICD2 in the two groups were significantly different (P=0.000);after treatment with NOTCH signaling pathway inhibitor S2188 for 48 h, the cell apoptosis rate was significantly increased (P=0.000);and the expression levels of Cleaved Caspase-3, HES1, NICD1 and NICD2 were significantly different between the two groups (P=0.000). Conclusion MTSS1 expression is down regulat-ed in gastric carcinoma. MTSS1 can promote the apoptosis of gastric cancer cells and inhibit the proliferation of gastric cancer cells, which is associated with the NOTCH signaling pathway.
