| 摘 要： | 目的探讨胃癌发病机制。方法应用巢式逆转录多聚酶链反应(NT-PCR)和免疫组织化学(PV-9000二步法)观察P53选择性剪接异构体与Bax、P21在胃癌和相应癌旁组织中的表达关系。结果在30对胃癌及其癌旁胃组织只检测到△133P53、P53βmRNA表达,而未检测到P53γ、△133P53β、△133P53γ。Δ133P53与Bax、P21呈负相关,P53β与Bax、P21呈正相关。结论胃癌组织中P53异构体的差异性表达通过与Bax启动子和P21启动子结合进而调节P53的活性,对胃癌的发生发展起了重要的作用。

| Abstract: | Objective To explore the mechanism of gastric carcinoma.Methods The expression of P53 alternative isoforms,Bax and P21 protein in gastric carcinoma and their corresponding tumor-adjacent tissues were detected using NT-PCR and immunochemistry methods.Results △133P53 and P53β were detected in both 30 cancer tissues and tumor-adjacent tissues.P53γ,△133P53β and △133P53γ were not detected in both 30 cancer tissues and tumor-adjacent tissues.Δ133P53 was negative correlation with both Bax and P21 protein.P53β was positive correlation with both Bax and P21 protein in gastric carcinoma.Conclusion The different expression of P53 isoforms with Bax and P21 protein plays an important role in the pathogenesis of gastric carcinoma by controlling P53 protein. |
