基于公共数据库分析CLDN10与MYH10在肾透明细胞癌中表达及预后的意义

摘    要:目的探讨CLDN10与MYH10在肾透明细胞癌中表达及预后的意义。方法通过GEO数据库分析获取肾透明细胞差异基因,并使用DAVID数据库对差异基因进行富集分析和功能注释,通过HPA数据库获取CLDN10与MYH10在正常肾脏组织蛋白表达图谱,再通过GEPIA数据库获取CLDN10与MYH10表达差异及肾透明细胞癌患者的预后信息。结果GEO数据库分析表达谱数据集GSE105261共得到438个差异基因,DAVID数据库富集分析表明这些差异基因主要富集在分泌过程、细胞外的外泌体、细胞外基质结构组分以及抗生素的生物合成。GEPIA数据库分析显示:CLDN10与MYH10在正常对照组中表达较肿瘤组显著增高(P<0.05),差异具有统计学意义。CLDN10表达与MYH10表达呈显著正相关(P<0.05),并且CLDN10、MYH10表达量与肾透明细胞癌患者的病理分期呈负相关(P<0.05),与总生存及无病生存呈明显正相关(P<0.05)。结论CLDN10与MYH10在肾透明细胞癌组织中低表达,并且与患者的生存预后明显相关,CLDN10与MYH10有望成为预测肾透明细胞癌患者预后的生物标志物及治疗的靶点。

Expression of CLDN10 and MYH10 and Their Prognostic Role in Clear Cell Renal Cell Carcinoma:an Analysis Based on the Data-mining of Public Database

Abstract:Objective To investigate the expression and prognostic significance of CLDN10 and MYH10 in clear cell renal cell carcinoma.Methods Differentially expressed genes in clear cell renal cell carcinoma(ccRCC)were obtained from the data set GSE66271 of GEO database.The DAVID database was used to analyze the functions of differentially expressed genes.The immunohistochemical results of CLDN10 and MYH10 in normal renal tissues were quoted from the HPA database,and patients prognosis of different expression of CLDN10 and MYH10 was analyzed by the GEPIA database.Results A total of 438 differentially expressed genes were obtained from the data set GSE66271 of GEO database.The enrichment analysis by the DAVID database indicated that these differentially expressed genes were mainly enriched in the excretion,extracellular exosomes,extracellular matrix structural constitution and biosynthesis of antibiotics.The GEPIA database analysis showed that the expression of CLDN10 and MYH10 in the normal group was significantly higher than that in the tumor group(P<0.05).CLDN10 expression was significantly positively correlated with MYH10 expression(P<0.05),and the expression levels of CLDN10 and MYH10 were negatively correlated with pathological stage of patients with ccRCC(P<0.05),and significantly positively correlated with overall survival and disease free survival(P<0.05).Conclusion CLDN10 and MYH10 are low expressed in ccRCC,and are significantly correlated with the survival and prognosis of patients.CLDN10 and MYH10 are expected to be biomarkers predicting the prognosis and therapeutic targets for patients with ccRCC.
